Multimodal single cell profiling of bone fracture microenvironment

Project description:Age-specific alterations of the adaptive immune system include low-grade chronic inflammation, which contributes to the unfavorable outcome of the bone healing process in the elderly. In this work, we aimed to characterize the effect of immune experience and age on cellular phenotype during the early inflammatory phase after bone fracture by using a mouse model which included young, aged, and immune-aged mice 2- and 5-days post osteotomy. Single-cell proteo-genomics quantified thousands of transcriptomes of cells isolated from three different bone regions (fracture hematoma, proximal and distal bone marrow). The fracture hematoma is a very distinct tissue consisting of coagulated blood caused by rupture of adjacent blood vessels, bone marrow influx and cells attracted from the surrounding tissue. The hematoma matures with the healing process and the adjacent bone marrow (proximal and distal to the fracture gap) supports the healing process.

Project description:Periosteum deficiency affects bone regeneration, especially in the case of the bone disorder congenital pseudarthrosis of the tibia (CPT). We investigated a new mouse model of CPT caused by Nf1 inactivation in boundary cap-derivatives, the Prss56Cre; R26tdTom; Nf1fl/fl model. To investigate altered healing in this CPT model, we generated a dataset of the injured periosteum and fracture callus at day-7 post fracture from Prss56Cre; R26tdTom; Nf1fl/fl mice.

Single nuclei RNAseq of the periosteal response to bone fracture in mice

Project description:Bone regeneration is a highly efficient process allowing scarless healing after injury. The periosteum, the outer layer of bones, is a critical source of skeletal stem/progenitor cells (SSPCs), as well as immune, endothelial and neural cells during bone repair. In our study, we generated a single-nuclei atlas of the murine periosteal response to bone fracture. We generated single nuclei datasets from uninjured periosteum and from injured periosteum and hematoma/fracture callus at days 5 and 7 post-injury from wild-type mice.

Project description:The specific genes that distinguish normal fracture healing from abnormal healing or nonunion in humans are unknown. This study was an exploratory investigation of peripheral blood from acutely injured fracture patients collected over multiple days compared to age- and sex-matched healthy volunteers. Bone fragments were collected from acutely injured subjects during surgery through the Cooperative Human Tissue Network-Eastern Division (CHTN-ED http://pathology.med.upenn.edu/research/centers-and-institutes/cooperative-human-tissue-network). We used microarrays to do a global comparison between acutely injured (AI) subjects (AIS) and chronic Nonunion (NU) subjects vs. age- and sex-matched healthy volunteers (HV). We also compared tissue types by capturing bone fragments from the fracture site where possible.

Yet, musculoskeletal traumatic injury, when fracture is combined with adjacent muscle injury, alters bone healing leading to fibrous nonunion. The periosteum, the outer layer of bones, is a critical source of skeletal stem/progenitor cells (SSPCs), as well as immune, endothelial and neural cells during bone repair. We generated single nuclei datasets from the injured perioteum and hematoma at day 1-post fracture and from the hematoma/callus at day 5 post-musculoskeletal traumatic injury. These datasets were analyzed in combination with datasets from GSE234451.

This study explored global RNA expression in remnant bone fragments from acutely injured patients undergoing open primary fracture repair surgery. These data will be compared to blood samples collected from acutely injured subjects, chronic nonunion subjects, and a panel of healthy volunteers. We used microarrays to do a global comparison across bone fragments collected from different patients, and different bones. We also evaluated the role of remnant bone preparation which was either immediate freezing in liquid nitrogen or heat stabilized with the Denator Stabilizer(TM) system.

Transcriptomic analysis of local bone tissue in rats after bone fracture

Project description:In this study, transcriptome sequencing technology was used to detect and analyze the changes in transcriptome of the local injury tissue after fracture. Differentially expressed genes (DEGs) with the high degree were analyzed. Our research results identify some important genes and pathways in the process of fracture healing, and summarize the main characteristics of transcriptome changes in fractures. This laid the foundation for a deeper understanding of the relation between molecular mechanisms and biological processes involved in fracture healing. By identifying the specific time points at which various cellular events occur during the healing process, researchers can gain insights into the complex interplay between cells, signaling pathways, and extracellular matrix components that is required for successful bone healing. This knowledge can inform the development of new therapies and interventions to improve fracture healing outcomes and ultimately enhance the quality of life for patients affected by bone fractures.

Proteomic analysis of successful and biologically compromised bone fracture healing

Project description:Bone fracture healing shows approximately 10-15% non-unions, although theoretically it has the potential of scarless regeneration. With regards to our aging society, a better understanding of the healing process is needed to allow for more sophisticated treatment options. Particular the early phase, which is a high complex and dynamic process in regards to nutrient and engery requirements. Here we investigate the proteomic (and metabolic) characteristics of the local microenvironment from successful (in young female Sprague-Dawley rats) and biologically compromised (in aged female rats with a minimum litter of three)bone reneration at day 3, 7 and 14 after osteotomy.
