Project description:Bronchopulmonary dysplasia (BPD) represents one of the most important sequelae of preterm birth affecting more than 20% of infants born below 32 weeks. Mechanical ventilation exerting shear stress (cyclic mechanical stress, CMS) and oxygen toxicity (hyperoxia, Hox) mainly contribute to the pathogenesis of BPD. The study proposal aims to separate the impact of CMS and Hox on pulmonary mesenchymal stem cells (MSC) isolated from preterm infants participating in the DZL PROTECT-AIRR cohort study in a world-wide unique in vitro setting. The impact of rhythmic 2D and 3D stretch to MSC cell cultures alone and in combination with different fractions of oxygen is evaluated with respect to MSC phenotype, transcriptional regulation, protein alterations and paracrine secretion.

Project description:Context: Increased uterine stretch appears to increase the risk of preterm labour, but the mechanism is unknown. Objectives: To identify a targetable mechanism mediating the effect of stretch on human myometrium. Design: Myometrial explants, prepared from biopsies obtained at elective caesarean delivery, were either studied acutely, or were maintained in prolonged culture (up to 65 h) under tension with either a 0.6 g or 2.4 g mass, and compared using in vitro contractility, whole genome array, and qRT-PCR. Results: Increased stretch for 24 or 65 h increased potassium-induced and oxytocin-induced contractility. Gene array identified 62 differentially expressed transcripts after 65 h exposure to increased stretch. Two probes for gastrin-releasing peptide (GRP), a known stimulatory agonist of smooth muscle, were among the top five up-regulated by stretch (3.4-fold and 2.0-fold). Up-regulation of GRP by stretch was confirmed in a separate series of 10 samples using qRT-PCR (2.8-fold, P = 0.01). GRP stimulated contractions acutely when added to freshly obtained myometrial strips in 3 out of 9 cases, but Western blot demonstrated expression of the GRP receptor in 9 out of 9 cases. Prolonged incubation of stretched explants in the GRP antagonists PD-176252 or RC-3095 (65 and 24 h respectively) significantly reduced potassium chloride and oxytocin-induced contractility. Conclusion: Stretch of human myometrium increases contractility and stimulates the expression of a known smooth muscle stimulatory agonist, GRP. Incubation of myometrium in GRP receptor antagonists ameliorates the effect of stretch. Project description:Preterm birth disrupts the critical maturation of the intestinal epithelium, leading to compromised digestive and absorptive functions. This study explores the potential of mesenchymal stem cell (MSC)-derived extracellular vesicles (EVs) to promote the maturation of human preterm intestinal epithelial cells. By establishing human preterm intestinal organoids derived from the ileal tissue of a neonate born at 27 weeks gestational age, we demonstrate that treatment with EVs, particularly the EV39 line, significantly enhances the expression of adult-type enterocyte markers, including genes involved in lipid and nutrient transport, digestive enzymes, and epithelial barrier integrity. These maturation signatures were identified through reanalysis of multiple single-cell RNA sequencing (scRNA-seq) datasets, revealing distinct enterocyte differentiation trajectories and key maturation-specific gene expression profiles. Notably, these maturation effects were dependent on cystic fibrosis transmembrane conductance regulator (CFTR) activity, as CFTR inhibition reversed the observed benefits. Proteomic analysis identified FGF2 and TGFβ1 as key ligands mediating the effects of EV39. Co-treatment with FGF2 and TGFβ1 further enhanced epithelial barrier integrity and fatty acid uptake, highlighting the translational potential of EV39 or its ligands in promoting intestinal functional maturation in preterm infants.

Project description:This study investigated how 14-day 4g hypergravity exposure affects mechanosensitive ion channels (MSCs) in rat ventricular cardiomyocytes. RNA-seq revealed selective MSC remodeling: TRPM7 (+41%), TRPC1 (+68%), and TRAAK (+239%) were upregulated, while TRPV2 (-62%), Piezo2 (-58%), and Kir6.1 (-76%) were downregulated. Structural analysis showed 44% increased membrane capacitance and sarcomere elongation, consistent with hypertrophic remodeling. Patch-clamp studies demonstrated markedly hypersensitive stretch-activated currents (I_SAC), with activation at minimal displacements (1-2 μm) and enhanced responses under 4-10 μm stretch (107-222% of controls). These coordinated molecular, structural, and functional adaptations suggest targeted transcriptional reprogramming that balances enhanced Ca²⁺ signaling with stabilized K⁺ conductance, demonstrating cardiac mechanotransduction plasticity under gravitational stress with implications for cardiovascular risks in altered gravity environments.
