In order to confirm which amino acid of the 3TD of SETDB1 is covalently modified by a set of cysteine-targeting ligands, we performed LC-MS/MS on sample containing 40 uM of SETDB1 3TD with 400 uM of ligand after overnight incubation at 4 C.
