Nonsense mutations generate premature termination codons (PTCs) that are responsible for 11% of human genetic disease alleles. Thus, nonsense suppressor tRNAs have broad therapeutic potential. Humans encode > 600 tRNA genes with many identical or similar copies of each tRNA. We hypothesized that tRN...

High-fidelity and differential nonsense suppression in live cells and a frontotemporal dementia allele with human transfer RNAs
Here, we report the discovery of a novel mechanism of genetic complementation in M. marinum. We found that nonsense suppression was causing genetic complementation. Nonsense suppression was linked to alterations in cell permeability and exposure to the aminoglycoside antibiotic, hygromycin.

Use of NanoLuc-Based Reporters that Monitor Nonsense Suppression and Inhibition of Nonsense-Mediated mRNA Decay to Identify Potent New Agents That Partially Reverse the Effects of Nonsense Mutations
A Ψ–Ψ codon–anticodon pairing in nonsense suppression and translational recoding
PTCs cause a multitude of human diseases and there are no established therapeutic options for their therapeutic management. Herein, we report the high-throughput cloning and identification, characterization and functional analysis of anticodon-edited tRNA which display efficacious PTC reversion in ...

Nonsense mutations generate premature termination codons (PTCs) that are responsible for 11% of genetic disease alleles. The arginine (Arg, CGA) to stop (UGA) mutation is the most common PTC. Humans encode >600 tRNA genes with many identical and similar copies. We developed a dual fluorescent report...
