We developed a tip-based sequential IMAC based on different kinds of metal ion for increasing the coverage of phosphoproteome. This approach not only can be used for singly and multiply phosphopeptides separation but also for acidic, basophilic and Pro-directed phosphopeptides separation.

Abstract: Despite recent advances in instrumentation and analytical strategies for identification and quantitation of protein phosphorylation, methodologies to enrich the heterogeneous types of phosphopeptides are critical towards comprehensive mapping of the under-explored phosphoproteome. Taking a...

Protein phosphorylation and N-glycosylation are key post-translational modifications (PTMs) in plants that regulate critical signaling processes. However, analyzing both PTMs simultaneously remains challenging due to low abundance and divergent enrichment requirements. Here, we present a single-tip ...

The field of phosphoproteomics has been revolutionized in the last years, with enhanced efficiency and sensitivity, now enabling deep phosphoproteomic analysis single-shot from minimal peptide inputs. In this study, we rigorously evaluate the effects of various experimental parameters in automated Z...

In this study we compared the phosphoproteome of control and cold acclimated Drosophila melanogaster in order to unravel the cold acclimation regulation mechanisms. For this prurpose we enriched phosphopeptides from each sample using a sequential elution from IMAC strategy (SIMAC) and analyzed them ...
