Project description:Mice (n=85) were anaesthetised by ip injection (0.1 ml/10 g) of xylazine (5%) and ketamine (10%) diluted in sterile water. After placing animals in a stereotactic frame, the skull surface was exposed and holes were drilled at the appropriate sites to allow bilateral infusion of ibotenic acid (10 ï¾µg/ï¾µl in saline) with a glass capillary (40 ï¾µm external tip) glued to a 1 ï¾µl Hamilton syringe connected to stereotactic syringe pump (KDS 310, KD scientific). Antero-posterior (AP), lateral (L) and vertical (V) coordinates (in mm) for micro-infusions in the dorsal hippocampus, were taken relative to the bregma: (1) AP -1.7, L +/- 1, V -1.5; (2) (2) AP -2.2, L +/- 1.5, V -1.5; (3) AP -2.6, L+/- 2, V -1.5 (based on brain atlas by Paxinos and Franklin, 2004). The infused volume of ibotenic acid or PBS was 30 nl per injection site. The rate of infusion was 30 nl/min and the needle was left in situ for another 2 min, before being slowly retracted.
Project description:Aortic samples ranging from the aortic valve to the diaphragm were harvested from 15 newborn and 15 six-week old C57/black6 wildtype mice. Frozen samples of 5 animals each were pulverized and directly dissolved in RNA pure (PeqLab) to get three samples for each group. Total RNA was isolated using phenol/chloroform extraction.<br><br>For gene-expression analysis, 500 ng total RNA of each RNA sample was labeled using the Agilent single-color Quick-Amp Labeling Kit and hybridized on Agilent Whole Mouse Genome Microarrays (4x44K). For<br><br>analysis of microRNA, 200 ng total RNA of each sample was labeled using the Agilent miRNA Labeling Reagent and Hyb Kit, and hybridized on Agilent Mouse miRNA Microarrays (8x15K) according to the manufacturer's instructions.

Project description:To identify genes and related pathways that are regulated by radiotherapy in acute lymphoblastic leukemia (ALL) patients lymphocytes, we have employed whole genome microarray expression profiling as a discovery platform to identify genes associated with total body irradiation (TBI) therapy. Purified peripheral lymphocytes were sampled 24 h after each 4.5 Gray TBI treatment (total 9 Gray) from 4 adult patients receiving hematopoietic stem cell transplantation, and the results showed that 478 genes were differentially expressed with increasing TBI dose, and serial test of cluster (STC) analysis suggested three major patterns in these genes. Modulated genes were constructed to co-expression network and identified significant GO as follow: ubiquitin-dependent protein catabolic process, transcription, proteolysis and apoptosis et al. The reliability of the data obtained from the microarray was verified by performing quantitative real-time PCR on 12 representative genes. This study provides candidate genes associated with TBI sensitivity for further research.
