Weight loss in visceral adipose tissue

ABSTRACT: The adipose progenitors and the macrophages were sorted from the epididymal adipose tissue of lean (8 weeks chow diet), obese (4 weeks HFD) and Formerly obese mice (4 weeks HFD and 4 weeks chow diet), and their transcriptomic profiles were performed using RNAseq.

Adipose tissue stromal cells from lean, obese, and formerly obese mice

Project description:Adipose tissue stromal cells contribute to the regulation of adipose tissue in lean and obese states. Myeloid cells such as adipose tissue macrophages (ATMs) and dendritic cells (ATDCs) undergo both quantitative and qualitative changes with obesity. Due to similarity in markers the identify of adipose tissue dendritic cells and macrophages has been elusive. We have refined prior protocols to unambiguously discern ATM and ATDC in mice. We used microarrays to compare the profiles of ATMs and ATDC from gonadal adipose tissue from lean, obese, and formerly obese mice. We also isolated preadipocytes (PA) from lean and obese mice for comparison. Male C57Bl/6 mice were fed normal diet (ND) or high fat diet (HFD) for 16 weeks. Weight loss (WL) mice were switched from the HFD to ND for 8 weeks. RNA was purified from FACS sorted cell populations (live cells only) obtained from gonadal/epididymal adipose tissue depots. ATMs were defined as CD11c+ (CD45+CD64+ CD11c+) or CD11c- (CD45+CD64+ CD11c-) ATMs. ATDC were defined as CD64- CD11c+. PA were defined as CD31- CD45- Sca1+ PDGFRA+.

A history of obesity leaves an inflammatory fingerprint in liver and adipose tissue [gWAT]

Project description:Dieting is a popular yet often ineffective way to lower body weight, as the majority of people regain most of their pre-dieting weights in a relatively short time. The underlying molecular mechanisms driving weight regain and the increased risk for metabolic disease are still incompletely understood. Here we investigate the molecular alterations inherited from a history of obesity. In our model, male HFD fed obese C57BL/6J mice, were switched to a low caloric chow diet, resulting in a decline of body weight to that of lean mice. Within seven weeks after diet switch, most obesity associated phenotypes, such as body mass, glucose intolerance and blood metabolite levels were reversed. However, hepatic inflammation, hepatic steatosis as well as hypertrophy and inflammation of perigonadal, but not subcutaneous, adipocytes persisted in formerly obese mice. Transcriptional profiling of liver and perigonadal fat revealed an upregulation of pathways associated with immune function and cellularity. Thus, we show that weight reduction leaves signs of inflammation in liver and perigonadal fat, indicating that persisting proinflammatory signals in liver and adipose tissue could contribute to an increased risk of formerly obese subjects to develop the metabolic syndrome upon recurring weight gain.

Project description:Ramulus Mori (Sangzhi) alkaloids (SZ-A) improves lipid metabolism and adipose tissue inflammation in HFD-induced obese mice.This study compares transcriptome profiling (RNA-seq) in the epididymal adipose tissue of normal chow, high-fat diet (HFD) control and SZ-A-treated HFD mice to verify the regulatory mechanisms of SZ-A. These results demonstrated that SZ-A regulates lipid metabolism and inflammation.

Weight loss resulted in increased expression of the anti-inflammatory protein, CRISPLD2, in mouse adipose tissue

Project description:Purpose: NGS was used to determine if a distinct transcriptomic profile is observed between lean, obese and weight loss fat Methods: We carried out RNA-Seq analysis of epididymal adipose mice ad libitum fed for 10 weeks either a high fat diet (HFD) or a regular chow diet (RD), versus a cohort of mice fed HFD for the first 5 weeks before swapping to RD for the remainder (SWAP). Results: SWAP feeding resulted in weight loss with a parallel improvement in insulin sensitivity. RNA-Seq revealed several transcriptomic signatures distinct to SWAP adipose, distinguished from both RD and HFD adipose. We found a unique up-regulated mRNA encoding a secreted, LPS-binding glycoprotein, CRISPLD2, in SWAP adipose tissue. While cellular CRISPLD2 protein levels were unchanged, plasma CRIPSLD2 levels increased in SWAP mice following weight loss, and can correlate with insulin sensitivity. Conclusions: Taken together, our data demonstrate that CRISPLD2 is a circulating adipokine that may regulate adipocyte remodeling during weight loss.

Proteomics analysis of insulin sensitive tissues in high fat diet fed mice

Project description:The popularity of high fat foods in modern society has been associated with epidemic of various metabolic diseases characterized by insulin resistance, the pathology of which involves complex interactions between multiple tissues such as liver, skeletal muscle and white adipose tissue (WAT). To uncover the mechanism by which excessive fat impairs insulin sensitivity, we conducted a multi- tissue study by using TMT-based quantitative proteomics. 3-week-old ICR mice were fed with high fat diet (HFD) for 19 weeks to induce insulin resistance. Liver, skeletal muscle and epididymal fat were collected for proteomics screening. Additionally, PRM was used for validating adipose differential proteins. By comparing tissue-specific protein profiles of HFD mice, multi-tissue regulation of glucose and lipid homeostasis and corresponding underlying mechanisms was systematically investigated and characterized. NC: normal birth weight + chow diet; NH: normal birth weight + high fat diet; LC: low birth weight + chow diet; LH: low birth weight + high fat diet.

Hepatic tissue expression profiles of insulin-sensitive obese mice, insulin-resistant mice and control mice

Project description:Male C57BL/6J mice were fed a high-fat diet (HFD, 60 kcal% fat, D12492, Research Diets, Inc) or normal standard chow diet with 10 kal% fat (ND, D09100304, Research Diets, Inc). Specifically, 6-week-old mice were fed a HFD for 12 weeks to induce insulin resistance (HFD-12w group); 14-week-old mice were fed a HFD for 4 weeks to induce obesity (HFD-4w group). Control mice were fed a ND continuously for 12 weeks starting at 6 weeks of age (ND group). All mice reached the experimental endpoint at 18 weeks of age. Insulin sensitivity was measured by glucose tolerance test and insulin tolerance test. Mice that developed insulin resistance in HFD-12w group and obese mice with normal insulin sensitivity in HFD-4w group were used for further experiments. Mice in ND group were used as controls. Upon reaching the experimental endpoint, livers from three insulin-resistant mice, three insulin-sensitive obese mice, and three control mice were removed for RNA sequencing.

LC-MS/MS of thoracic aorta and perivascular adipose tissue following short-term dietary methionine restriction

Project description:We are exploring pathways influenced by short-term dietary methionine restriction (MR) in thoracic aorta and associated perivascular adipose tissue (PVAT) from obese C57BL/6J male mice around 26 weeks of age. Mice were fed 60% high fat diet (HFD) until they were obese, experimental mice were given 60% HFD with 0.12% methionine for 3, 5, and 10 days while the control group maintained a 60% HFD with 0.86% methionine. All mice were euthanized on the same day, then thoracic aorta was harvested, the PVAT was removed, and both sample types were prepared and analyzed separately. Three mice per group were assessed with LC-MS/MS. MR diet induced lean phenotype in PVAT, which was observed histologically, and we hypothesized that changes in lipid metabolism and mitochondrial bioenergetics mediated this response.
