ABSTRACT: Primary human monocytes were isolated and differentiated into moDCs. These moDCs were infected with WT Dengue virus 2 and its attenuated mutants (generated using infectious clone construction with site-directed mutagenesis) to better understand the gene expression changes induced by these different attenuated viruses.

Project description:Daily transcriptomic profiling was conducted on whole blood collected from COVID-19 cases. Whole blood was collected in Tempus Blood RNA tubes, and RNA was extracted from whole blood using the Tempus Spin RNA Isolation Kit. Healthy controls consisted of participants of a measles, mumps and rubella re-vaccination study. Pre-vaccination whole blood was collected and processed and analyzed as above.

NanoString profiling of host response in healthy volunteers vaccinated with ARCT-021 self-amplifying mRNA vaccine or placebo

Project description:Transcriptomic profiling was conducted on whole blood collected from study participants (NCT04480957). A total of 106 healthy participants were randomized into single and two-injection cohorts. Gene transcript levels were determined at baseline (pre-dose 1), day 2, 3, and 8 for all vaccinees. In addition, gene expression was profiled at day 29 (pre-dose 2), 30 and 36 among vaccinees in the two-injection cohort. Whole blood was collected in Tempus Blood RNA tubes, and RNA was extracted from whole blood using the Tempus Spin RNA Isolation Kit.

The RNA from 0h-post LPS stimulation were used as reference sample. Were also performed dye-swaps of each biological sample as technical replicates.

Project description:The transcription factor β-catenin has been shown to be active in different types of dendritic cells (DCs) with ability to induce tolerogenic or anti-inflammatory features. Monocyte-derived dendritic cells (moDCs) have been widely used in dendritic cell-based cancer therapy, but so far with limited clinical efficacy. It is possible that aberrant differentiation or induction of dual pro- and anti-inflammatory features may decrease the moDCs efficiency to stage the immune attack on cancer cells. Here we show, using moDCs derived from healthy buffycoats, that β-catenin is detectable in both immature and lipopolysaccharide (LPS)-matured DCs.
