LincK promotes proliferation and epithelial-to-mesenchymal transition and contributes to tumorigenesis and growth in breast cancer II

ABSTRACT: We previously reported that transwell co-culture with hAD-MSCs cells can induce an EMT progress in MCF7 cells. To identify EMT-relevant lncRNAs, we conducted transcriptome microarray analysis of MCF7 cells at Day 0, 2, 4, 6, 8 and 10 after coculture.

Project description:In this study, we aimed to identify potential lncRNA deregulations associated with breast cancer malignancy instigated by MSCs-MCF7 co-culture. We profiled expression changes of lncRNAs in MCF-7 cells during EMT induced by coculture with hAD-MSCs, and found an intergenic lncRNA with proviouly unknown function (KB-1732A1.1, we termed it LincK), which was significantly elevated. Depletion of LincK decreased the growth, migration, invasion, and EMT in breast cancer cells, while overexpression of LincK exerted the opposite effects. Moreover, knockdown of LincK repressed tumorigenesis, and ectopic expression of LincK promoted tumor growth in MCF-7 xenograft model. LincKs can act as a sponge of mirR-200b, which inhibits its function in proliferation and metastasis. Thus we reported, for the ﬁrst time, the role of LincK in control of EMT and proliferation in breast cancer. We used microarrays to detail the genes associated with LincK.

Project description:The epithelial-to-mesenchymal transition (EMT) process in cancer enables the migratory and invasive capabilities associated with metastatic competence. The cancer microenvironment plays an important role in inducing the occurrence of EMT. A number of extracellular stimuli and associated signaling pathways have been identified to promote EMT, however, molecular features during the early stages of EMT are poorly understood. Here we have established a novel in vitro EMT-inducing system that mimi cs tumor microenvironment and allows for close monitoring of the EMT progress. Using RNA-seq and miRNA-seq, we analyzed the transcriptomic transitions accompanying the progression of EMT, and pinpointed several molecular features during early EMT of the lung carcinoma cells.

Adipose MSC suppress breast cancer progression, metastasis and inhibit EMT pathways via exosomal-miRNAs following co-culture interaction

Project description:Introduction: Mesenchymal stem cells (MSCs) are commonly known to influence the progression of cancer due to their ability to migrate to the tumor microenvironment and interact with cancer cells. Exosomes have been found secreted by most cell types and have been shown to act as cargo carrying biomolecules including microRNA (miRNAs). Thus, understanding interaction between MSCs and cancer cells via exosomal miRNAs is crucial in determining the therapeutic role of MSC in treating breast cancer cells and relapse. Methods: Exosomes were harvested from the medium of indirect co-culture of MCF7-luminal and MDA-MB-231-basal breast cancer cells (BCCs) subtypes with adipose MSCs and profiled for miRNAs using next-generation sequencing. Results: The interaction resulted in the changes of exosomal miRNAs profiles that modulate essential signalling pathways and cell cycle arrest into dormancy via inhibition of epithelial-to-mesenchymal transition (EMT). The maintenance and induction of epithelial features in MCF7 and MDA cells have led to a positive correlation with the reduction of proliferation and metastasis as well as increased drug resistance. Overall, adipose MSCs mediated delivery of consensus miRNAs particularly miR-200 family in MCF7, miR-146a in MDA and miR-941 in both BCCs subtypes via exosomes.

Project description:To identify miRNAs participating in SNAI1-orchestrated regulatory pathways, we analysed time-resolved microarray data of SNAI1-induced EMT, obtained during conditional expression of SNAI1 in a “Tet-Off” MCF7-SNAI1 breast carcinoma cell model (Vetter et al, 2009). miRNA time series study for 7 times points (4h, 8h, 12h, 24h, 48h, 72h, 96h) in 3 replicates. For each time point, we compared induced and non-induced samples. Overall, we have 42 samples (21 hybridizations).
