Transcriptome sequencing analysis for NCI-H209 lung tumor cells conditioned by human brain microvascular endothelial cells

ABSTRACT: Endothelial cells of blood vessel playing a key role in the metastasis of tumor cells particularly the brain microvascular endothelial cells in the brain-tropic metastasis of lung tumor cells. Currently, studies towards the influence contributed by tumor cells to the alteration of endothelial cells have been extensively conducted by contrast studies for the alteration of gene-expression signature of tumor cells under the influence of endothelial cells remains poorly characterized, thus in this study the transcriptome of the human small-cell lung cancer cell NCI-H209 was sequenced via RNA-Seq after conditioned by human brain microvascular endothelial cell (HBMEC) for characterizing the alteration of gene-expression signature.

Plasmodium falciparum and TNF-a Differentially Regulate Inflammatory and Barrier Integrity Pathways in Human Brain Endothelial Cells

Project description:Cerebral malaria is a severe complication of Plasmodium falciparum infection characterized by the loss of blood-brain barrier (BBB) integrity, which is associated with brain swelling and mortality in patients. P. falciparum-infected red blood cells and in!ammatory cytokines, like tumor necrosis factor alpha (TNF-a), have been implicated in the development of cerebral malaria, but it is still unclear how they contribute to the loss of BBB integrity. Here, a combination of transcriptomic analysis and cellular assays detecting changes in barrier integrity and endothelial activation were used to distinguish between the effects of P. falciparum and TNF-a on a human brain microvascular endothelial cell (HBMEC) line and in primary human brain microvascular endothelial cells. We observed that while TNF-a induced high levels of endothelial activation, it only caused a small increase in HBMEC permeability. Conversely, P. falciparum-infected red blood cells (iRBCLs) led to a strong increase in HBMEC permeability that was not mediated by cell death. Distinct transcriptomic pro"les of TNF-a and P. falciparum in HBMECs con"rm the differential effects of these stimuli, with the parasite preferentially inducing an endoplasmic reticulum stress response. Our results establish that there are fundamental differences in the responses induced by TNF-a and P. falciparum on brain endothelial cells and suggest that parasite-induced signaling is a major component driving the disruption of the BBB during cerebral malaria, proposing a potential target for much needed therapeutics.

Marinobufagenin regulates gene expression in human brain microvascular endothelial cells in vitro.

Project description:To understand the mechanism by which MBG increases permeability of human brain microvascular endothelial cell (HBMEC) monolayers, the primary cultures were treated with 10nM MBG for 12 h. RNA was isolated and analysed for gene expression on Agilent one color microarrays. Triplicate wells of HBMEC were treated with 10 nM MBG in DMSO or with just the drug vehicle for 12 h. RNA was isolated from each well and used to reverse transcribe and generate Cy3-labelled cRNA. Microarrays were hybridized, washed and scanned. Data were normalized and assessed for differential gene expression between MBG treated and control HBMEC.

Project description:We describe here the generation and characterization of immortalized HCPEnC (iHCPEnC) by ectopic expression of hTERT in primary cells using a lentiviral vector system without additional support by a viral oncogene. RNA sequencing (RNA-seq) analysis revealed that iHCPEnC retained an RNA expression profile very similar to the primary HCPEnC and different from that of other endothelial cell types as Human Brain Microvascular Endothelial Cells (HBMEC) and Human Umbilical Vein Endothelial Cells (HUVEC).

S.suis has evolved precise mechanisms to alter gene expression depending on the distinct challenges posed by particular disease sites. Herein, a whole-genome DNA microarray was used to investigate the change of gene expression profile of S.suis after contact with hBMEC 3 h. comparison of RNA isolated from hBMEC-associated S.suis with RNA derived from control bacteria revealed significant differential changes for 175 S.suis genes including 123 up-regulated genes and 52 down-regulated genes at 3 h post infection. A cDNA microarray imprinted with 2156 genes representing about 98% of Streptococcus suis serotype 2 genome was used for transcriptome analysis . For two-sample (reference vs. test) microarray hybridization, Two-condition experiment, control vs contact with hBMEC 3 h, four replicates (two biological replicates, two technical replicates) at each condition.
