ABSTRACT: Prkar1a encodes the regulatory subunit R1a of the proteine kinase A. Prkar1a ablation in the testis results in profound alterations of tissue homeostasis leading to tumor development and infertility. We used microarrays to analyze gene expression in response to Prkar1a gene ablation in testicular somatic cells (derived from SF1+ cells) (prKO model), in Sertoli cells (sKO model) or in Leydig cells (lrKO model) in 2-week and 2-month-old mouse testes.

Project description:We demonstrate that PKA signalling drives zonal conversion within adult adrenocortical lineage in a sexually dimorphic manner. Our data establish that Prkar1a genetic ablation (leading to constitutive PKA activation) in the adult adrenocortical lineage leads to endocrine hyperactivity and accelerates adrenal cortex renewal. This results in increased zona fasciculata differentiation and final conversion into reticularis-like zone. This phenomenon relies partly on sex-dependent mechanisms of cortical renewal, on which the male androgenic milieu exerts a repressive action through induction of WNT signalling, which in turn antagonizes PKA signalling and cortical cell turnover. We used microarrays to analyse gene expression in response to Prkar1a gene ablation in the adult adrenocortical cell lineage in both female and male mice and explored the effects of orchidectomy (GDX).

Project description:We wished to investigate the role of E-cadherin loss in our mouse parietal cell/pre-parietal cell E-cadherin knock-out, p53 knock-out, oncogenic Kras induced model of gastric cancer. As such, we isolated RNA from stomach tissue from our E-cadherin knock-out model (Atp4b-Cre;Cdh1(fl/fl);Kras(LSL-G12D/+);Trp53(fl/fl);Rosa26(LSL-YFP/LSL-YFP)) and our E-cadherin heterozygous model (Atp4b-Cre;Cdh1(fl/+);Kras(LSL-G12D/+);Trp53(fl/fl);Rosa26(LSL-YFP/LSL-YFP)). We then performed a microarray on this stomach tissue from four independent mice of each genotype. Differentially expressed genes were identified and gene set overlap analysis was used to identify pathways enriched in one model over the other.
