In vivo CRISPR activation screening of tumor formation in murine pancreas and lung.

ABSTRACT: We performed in vivo CRISPR activation screening of tumor formation in the pancreas and lung by treating CRISPRa competent PPKS (KRAS(LSL-G12D//wt); TP53(flox/flox); Rosa26(LSL-SAM/wt)) mice with CMV-Cre/U6-sgRNA(MS2) lentivirus containing the Onco1 sgRNA guide library. Tissues were harvested at different timepoints (6 days, 4 weeks, 8 weeks and survival endpoint). HEK293T cell cultures were transduced with the same libraries to generate normalization controls. Library plasmid DNA (pDNA) was sequenced as controls.
