Gene expression analysis of cancer-associated fibroblasts from scirrhous gastric cancers and non-scirrhous gastric cancers

ABSTRACT: Scirrhous gastric cancer has aggressive clinical courses and is characterized by dominating cancer-associated fibroblasts (CAFs). We here aimed to explore the molecular basis for scirrhous CAFs and their origin. The aggressive phenotype of scirrhous CAFs were associated with activation of TGFβ1 and NF-kB, along with increased expression of their downstream genes.

Twist1 knockdown in two gastric cancer-associated fibroblast lines

Project description:Primary human gastric cancer-associated fibroblast (CAF) cultures (CAF14 and CAF32) were establised from 2 gastrectomy specimens. Silencing the expression (loss-of-function effect) of Twist1 in CAFs showed candidate target genes regulated by Twist1 and abrogated their tumor-promoting properties. Experiment design: two gastric CAFs (CAF14 adn CAF32, respectively) were establised from two gastric cancer patients and Twist1 knockdown studies were performed using shRNA.

Syndecan-4 is a maestro of gastric cancer cell invasion and communication that underscores poor survival

Project description:Gastric cancer is a dominating cause of cancer-associated mortality with limited therapeutic options. Here, we show that syndecan-4 (SDC4), a plasma membrane proteoglycan, is highly expressed in intestinal subtype gastric tumours and that this signature associates with patients’ poor survival. Further, we mechanistically demonstrate that SDC4 is a master regulator of gastric cancer motility and invasion. We also find that SDC4 conjugated with heparan sulfate chains is efficiently sorted in extracellular vesicles (EVs). Interestingly, SDC4 in EVs regulates gastric cancer cell-derived EV organ distribution, uptake and functional effects in recipient cells. Specifically, we show that SDC4 knockout disrupts the tropism of EVs for the common gastric cancer metastatic sites. Our findings set the basis of the molecular implications of SDC4 expression in gastric cancer cells and open new perspectives on the development of therapeutic strategies targeting the glycan-EV axis to limit tumour progression.

Chemotherapy-induced senescent fibroblasts promote NF-kB-mediated tumor aggressiveness, suppressed by Rofecoxib in gastric cancers

Project description:Purpose: Chemotherapies for patients with gastric cancers (GCs) has been shown disappointing results in the clinics. The cancer-associated fibroblasts (CAFs) could be considered one cause of the resistance to chemotherapy in GCs. However, the chemotherapy-induced reprogramming of CAFs and its effects on GC progression and chemotherapy resistance have not been elucidated. Methods: IncuCyte and transcriptome were used to identify chemotherapy-induced senescence of CAFs. Screening for drugs to inhibit the senescence of CAFs was performed. The cell proliferation and migration assays and phospho-protein array were performed to investigate the effects of senescent CAFs on GC cells. Inhibitory effects of screened drugs on CAFs were also evaluated in experimental models. Senescent CAFs and their clinical meanings were also investigated in the human GC tissues undergoing preoperative chemotherapies. Results: Chemotherapy could alleviate proliferation and widen cytoplasm of CAFs. Some secretory molecules like TNF-α were upregulated in the CAFs with chemotherapy. Chemotherapy-treated CAFs increased the resistance to chemotherapy and the migratory potentials of GC cells. The COX2 inhibitor, one of the screened drugs, has inhibitory effect on chemotherapy-induced senescence CAFs. Consequently, it could suppress GC progression and resistance to chemotherapy. The GC patients with senescent CAFs were significantly correlated with poor histologic response to chemotherapy. Conclusions: In this study, we are convinced that the chemotherapy-induced senescence of CAFs could have a significant effect on progression and resistance to chemotherapy in GC. Inhibitory drugs for chemotherapy-induced senescence of CAFs could be used to overcome the CAF-induced resistance to chemotherapy in GCs.

RNA sequencing of cancer-associated fibroblasts (CAFs) and normal fibroblasts from a triple-transgenic gastric cancer mouse model

Project description:Gastric cancer is the 3rd most lethal cancer worldwide, and the genomic status of its cancer cells has not translated into effective prognostic or therapeutic strategies. We therefore hypothesize that outcomes may depend on the tumor microenvironment (TME), and in particular cancer-associated fibroblasts (CAFs). However, very little is known about the role of CAFs in gastric cancer. To address this, we map the transcriptional landscape of human gastric cancer stroma by microdissection and RNA sequencing of CAFs from gastric cancer patients. We find a stromal gene signature associated with poor disease outcome. This signature is regulated by the transcription factor Heat Shock Factor 1 (HSF1), that activates CAF-derived extracellular vesicles (EVs) that transfer components including Inhibin Subunit Beta A (INHBA) and Thrombospondin 2 (THBS2), to the TME to promote cancer. Together, our work provides the first transcriptional map of human gastric cancer stroma, and highlights HSF1 and its transcriptional targets as potential diagnostic and therapeutic targets in the genomically stable tumor microenvironment.

Fibrous stromal component in hepatocellular carcinoma reveals a cholangiocarcinoma-like gene expression trait and epithelial-mesenchymal transition.

Project description:Hepatocellular carcinoma (HCC) and cholangiocarcinoma (CC) are the major primary liver cancers in adults. It has been shown the phenotypic overlap between HCC and CC, comprising continuous liver cancer spectrum. As a proof of concept, a recent study has demonstrated a genomic subtype of HCC expressing CC-like gene expression trait, i.e. CC-like HCC, which has revealed common genomic trait of stem cell-like property and aggressive clinical outcome. However, the histopathological characteristics of these intermediate phenotype was not fully evaluated yet. Here, we found that a variant HCC with fibrous stromal component, i.e. scirrhous HCC, has CC-like genomic features. By performing gene expression profiling and immunohistochemical evaluation, we compared the morphological and molecular features of scirrhous HCC with CC and classical HCC. We found that the scirrhous HCC express both CC-like and stem cell-like expression traits. In addition, we observed the expression of core epithelial-mesenchymal transition (EMT)-related genes, which may contribute to the aggressive behavior of scirrhous HCC. Over-expression of transforming growth factor Î² (TGF-Î²) signaling was also found, implying its regulatory role in the pathobiology of scirrhous HCC. Conclusion: We suggest that the fibrous stromal component in HCC may contribute to the acquisition of CC-like gene expression trait in HCC. The expression of stem cell-like trait and TGF-Î²/EMT molecules may play pivotal roles in the aggressive phenotype of scirrhous HCC Gene expression profiles of HCC, CC and HCC with fibrous stroma were compared.
