Pseudomonas isolates reconfigure gene expression in walnut seedlings exposed to salt stress

ABSTRACT: To understand the potential mechanisms underlying the effects of the four Pseudomonas strains P1, P2, P3, and P4 on the growth and salt tolerance of walnut trees, the transcriptomic responses of walnut seedlings inoculated with these isolates under conditions of salt stress were further evaluated using RNA-seq analysis.

Culture adaptation occurs in hESCs during repeated in vitro culture to acquire ‘survival advantage’ to be highly resistant to various stresses. In special, difference in gene expression profile of cell death or apoptotic gene signature was evident between P1/P2 and P3/P4 hESCs.

Project description:To characterize the sequence of events associated with RasV12 immortalization of Drosophila embryonic cells, we generated transcriptional time series during cell line establishment, from primary cultures until passage (P) 19. We generated three transcriptional time series from three cell lines (R1, R4 and R5) by sampling the cultures at successive stages, early (P2-4), intermediate (P4-11), and late (P16-19), characterized by different passage times. Time points for the R1 time-series were: P2, P3, P4, P5, P7, P8, P10, P11, P16, P17 and P19; for the R4 time-series: P2, P3, P4, P5, P6, P7, P9, P11, P12, P16, P17 and P19; and for the R5 time-series: P2, P3, P4, P6, P7, P8, P16, P17 and P19

Characterization of CD44low CD62Llow CD8 T-cell subset

Project description:To characterize CD44low CD62low CD8 T-cell subset (P4), we compared gene expression among four CD8 T-cell subsets. A hierarchical clustering of global gene expression showed that each subset of CD8 T-cells expresses a distinct gene profile. Genes expressed in activated T-cells were highly expressed in P4 cells compared to naive P1 cells. On the other hand, genes expressed in differentiated T-cells were less expressed in P4 cells compared to either central memory (P2) or effector/memory (P3) cells. These results indicate that P4 cells are in an activated, but pre-mature state and distinct from more differentiated subsets of CD8 T-cells such as central memory and effector/memory cells.

Proteomic Characterization and Antiproliferative Potential of Proteins from Cuscuta Epithymum Crude Herbal Product Against MCF-7 Human Breast Cancer Cell Line

Project description:Phytochemicals from Cuscuta have been reported for exhibiting anticancer properties; however, the bioactivities attributed to the proteins remained unexplored. This study examined the proteins from a crude herbal product of Cuscuta epithymum (CE) harvested from host plants Alhaji maurorum and Medicago sativa for inhibiting the proliferation of the MCF-7 breast cancer cell line. The CE crude proteins (CP) were separated into four protein peaks (P1, P2, P3, and P4) by gel filtration chromatography. CP, P1, P2, and P3 showed dose-dependent growth inhibitory response in the MTT assay with respective IC50 values of 33.8, 43.1, 34.5, and 28.6 µg/ml. In western-blot analysis, upregulation of BCL2-associated X protein (BAX) with concomitant downregulation of BCL-2 in the cells treated with crude and P3 was observed. In cells treated with P2, BAX was upregulated; however, no significant change in the expression of BCL-2 was recorded. The LC-MS/MS analysis revealed the identity of those proteins in CE, such as peroxidase, non-specific lipid transfer protein, and superoxide dismutase, etc., which have been previously reported from other plants for exhibiting anticancer activity. The findings indicated that Cuscuta proteins strongly inhibited the proliferation of breast cancer MCF-7 cells by apoptosis. The survey of the CE protein profile suggests it as an enriched source of proteins with high biological significance.

Monitoring of functional gene responses to ERD (enhanced reductive dechlorination) from four TCE-contaminated sites

Project description:A functional microarray targeting 24 genes involved in chlorinated solvent biodegradation pathways has been developed and used to monitor the gene diversity present in four trichloroethylene (TCE) contaminated sites under ERD (enhanced reductive dechlorination) treatment. The microarray format provided by NimbleGen and used in this study is 12x135K. 2 M-BM-5g of labelled gDNA from 30 groundwater samples were hybridized on the microarrays. A 30-chip study was performed, each chip corresponding to hybridization with 2 M-BM-5g of labelled gDNA retrieved from a monitoring well from one of the four contaminated sites. Each probe (760nt) on the microarray was synthesized in eight replicates, and a total of 5,707 random probes was used to determine the background noise. Groundwater samples were collected from four contaminated sites (B, F, G and H), four monitoring wells per site (P1, P2, P3 and P4). P1: well located upstream to the contamination source. P2: well in the contamination source. P3 and P4: wells located downstream to the contamination source. For site B, the monitoring of ERD demonstration was performed through a total of 5 sampling campaigns: C1 (T=0), C2 (T=104 days), C3 (T=231 days), C4 (T=291 days) and C5 (T=378 days). For the three other sites (F, G and H), only one sampling campaign was performed after the treatment.
