Single-cell RNA sequencing of liver CD45+ immune cells in mouse acute-on-chronic liver failure

ABSTRACT: Single-cell RNA sequencing was performed on liver CD45+ immune cells isolated from mice subjected to acute-on-chronic liver failure (ACLF). Three experimental groups were included: CCl4-treated control mice, ACLF mice treated with vehicle, and ACLF mice treated with the EP300 inhibitor CCS1477. Male C57BL/6J mice were intraperitoneally injected with carbon tetrachloride (CCl4, 0.2 ml/kg, twice weekly) for 8 weeks. ACLF was induced by administration of a double dose of CCl4 (0.4 ml/kg), followed by intraperitoneal injection of Klebsiella pneumoniae. CCS1477 was administered intraperitoneally 2 hours before bacterial challenge. Liver CD45+ immune cells were isolated 36 hours after K. pneumoniae injection and subjected to BD Rhapsody whole-transcriptome single-cell RNA sequencing.
