Integrated Multi-Omics Analysis Links GAD1 Overexpression to Purine Metabolic Reprogramming in Lung Adenocarcinoma

ABSTRACT: RNA sequencing was performed to characterize transcriptomic changes associated with GAD1 overexpression in A549 lung adenocarcinoma cells. A549 cells transduced with GAD1-expressing lentiviral particles (adGAD1) were compared with A549 cells transduced with GFP control lentiviral particles (adGFP), with three biological replicates per group. Gene-level expression profiling was used to identify transcriptional changes and pathways associated with GAD1 overexpression.

Î±-catulin play a role in lung cancer metastasis

Project description:Background: Î±-catulin may functions as an oncoprotein, sustaining proliferation by preventing cellular senescence and promoting cancer cell migration. In this study, we investigated the mechanism of Î±-catulin in cancer cell migration and metastasis in lung cancer. Method: Î±-catulin mRNA expression was isolated from A549/AS2neo (control) and A549/AS2neo-Î±-catulin stable cells. The Phalanx Human OneArray microarray analysis was performed to identify Î±-catulin downstream genes. Results: Overexpression of Î±-catulin increased cancer cell migration and metastasis. By using Phalanx Human OneArray microarray we have identified panel of genes altered by Î±-catulin overexpression, consisting of CDC42, intergrins and genes related to cytoskeleton remodeling. Conclusion: Î±-catulin is an oncoprotein and promotes lung cancer cell migration and metastasis. Two-condition experiment, Vector vs. alpha-catulin overexpression cells.The cDNAs encoding full-length human alpha-catulin were amplified and subcloned into lentiviral pLKO_AS2.neo which generated full-length alpha-catulin. Vector control or alpha-catulin lentivirus were transduced into A549 cells and G418 was used to select stable cells.

The role of the transcription factors GATA2 and FOXA1 in immortalized basal-like prostate epithelial cells

Project description:GATA2 and FOXA1 are pioneering factors for Androgen Receptor (AR) in prostate cancer cells. Less is known about their role in benign epithelial prostate cells. We investigated if they had the ability to induce differentiation in an undifferentiated prostatic context. Benign basal-like prostate epithelial cells 957E/hTERT-AR cells (a gift from John T. Isaacs lab, Johns Hopkins, MD, USA) were transduced with LentiORF RFP control lentiviral particles. The 957E/hTERT-AR-RFP cells were then transduced with GATA2 and FOXA1 lentiviral particles, and we called the new cell lines hTERT/AR-GATA2 and hTERT/AR-FOXA1, respectively. hTERT/AR-GATA2 is a stable selected cell line and treated with 1 nM anabolic steroids, R1881, or ethanol (ET-OH) for 48 hrs. 957E/hTERT/AR-FOXA1 is a transient cell line, because FOXA1 protein is quickly lost. Twenty four hours post transduction with FOXA1 lentivral particles, 1 nM androgen R1881 or ethanol (ET-OH) were added at time for medium exchange and further treated for 24 hours. GATA-2 and FOXA1 ability to induce differentiation was assessed.

We used microarrays to analyze changes in A549 lung cancer cell line in which ALDH activity was reduced using lentiviral mediated expression of siRNA against both isozymes (Lenti 1+3) Experiment Overall Design: A549 lung cancer cell lines were transduced with lentiviral vectors containing specific siRNA sequences against ALDH1A1, ALDH3A1, both vectors (Lenti 1+3 cells), and against the green flourescent protein (GFP) gene (GFP cells, used as control).

Project description:We examined the transcriptional changes modulated by estrogen receptor beta (ERβ) by performing global transcriptome analysis. U87 cells were transduced with lentiviral particles carrying either empty vector or ERβ-FLAG expression vector and the RNA was isolated and utilized for RNA-seq analysis. Our results demonstrated that ERβ modulated genes were related to homologous recombination, DNA damage response, ATM signaling and cell cycle pathways.

In vitro expression data from Prdm16 overexpression in Blood Outgrowth Endothelial Cells (BOECs)

Project description:Previous work of the host lab identified Prdm16 as an arterial-enriched transcription factor (Aranguren et al., Blood 2013). To expand our knowledge on the genes regulated by Prdm16, we performed an Affymetrix microarray on total RNA of BOECs. These BOECs were transduced with Prdm16-overexpressing or Cherry-control lentiviral particles. Three replicate samples were subjected to microarray processing and Prdm16 samples were compared with Cherry control samples for further analysis.
