Comprehensive Immunoproteome of Mycoplasma bovis

ABSTRACT: This study was aimed to elucidate a global antigenic profile of Mycoplasma bovis (M. bovis) with immunoproteomics, immunoinformatics, and gene expression approaches. The extracts of whole-cell proteins and TX-114 membrane fraction of a Chinese strain M. bovis HB0801 were separated with two dimensional gel electrophoresis (2-DE) and proteins reacting with antisera to M. bovis from experimentally infected calves were detected by MALDI-TOF MS.

A lack of knowledge regarding the antigenic properties of Mycoplasma bovis proteins prevents the effective control of bovine infections using immunological approaches. Early events on compatible and incompatible grapevine interaction with Plasmopara viticola: a proteomic perspective of cultivar specific behavior

Project description:Grapevine downy mildew is an important disease affecting crop production and causing severe losses. To identify genotype-dependent responses towards this pathogen and to explore the molecular mechanisms involved in grapevine-P. viticola resistance, we have conducted a proteomic analysis of leaf samples from resistant and susceptible grapevine genotypes prior and post-inoculation with the pathogen. Proteins were analyzed by quantitative two-dimensional differential gel electrophoresis (2D-DIGE). The analysis able to identified 50 unique proteins. Functional analysis showed that photosynthesis and metabolism were the main categories differentiating genotypes at 0h and that P. viticola-responsive proteins were mainly involved in photosynthesis, carbohydrate metabolism, stress and defense responses and redox homeostasis. ROS production, total antioxidant capacity and lipid peroxidation on both genotypes were determined and together with the proteome data suggest that Regent presents a strict balance between ROS control and signaling leading to plant cell death activation. Our data reveals the genotype-dependent modulation of plant metabolism and defense responses providing new insights into underlying molecular processes of grapevine resistance against the downy mildew fungus.

Characterization of loquat (Eriobotrya japonica Lindl.) honey botanical origin according to its chitinase activity

Project description:Bees make honey from the nectar that they collect from flowers. The characteristics of honey are closely associated to original botanical species. Compare with sugars in honey, proteins are minor components but usually used as an important honey quality evaluation parameters. Flower-origin proteins could be a good marker for the authentication. However, as a minute component in honey proteome, plant origin proteins are hard to be detected in honey by regular proteomic approaches, such as gel-based techniques. In this study, Eriobotrya japonica Lindl. (loquat) nectar and its derivative monofloral honey were systematically compared, especially regarding the proteomes and enzymatic activities. Using two-dimensional electrophoresis and mass spectrometry, only bee-originated proteins were detected in loquat honey which were major royal jelly proteins and two uncharacterized proteins. Xylosidase, thaumatin, and two kinds of chitinases were detected in loquat floral nectar by the gel-based proteomic approach. To our knowledge, it is the first study to analysis nectar-originated enzymes’ activity in honey and we proposed that the zymography of chitinase is a potential marker for honey botanical origin authentication.

Characterization of a L-Gulono-1,4-Lactone Oxidase Like Protein in the Floral Nectar of Mucuna sempervirens, Fabaceae.

Project description:Floral nectar plays important roles in the interaction between animal-pollinated plants and pollinators. Growing empirical evidence shows that most of the proteins secreted in nectar (nectarins) are enzymes that can tailor nectar chemistry for their animal mutualists or reduce the growth of microorganisms in nectar. However, to date, the function of many nectarins remains unknown, and very few plant species have had their nectar proteome thoroughly investigated. Mucuna sempervirens (Fabaceae) is a perennial woody vine native to China. Nectarins from this species were separated using two-dimensional gel electrophoresis, and analyzed using mass spectrometry. A L-gulonolactone oxidase like protein (MsGulLO) was detected. MsGulLO has high similarity to L-gulonolactone oxidase 5 (AtGulLO5) in Arabidopsis thaliana, which was suggested to be involved in the pathway of ascorbate biosynthesis; however both MsGulLO and AtGulLO5 are divergent from animal L-gulonolactone oxidases. MsGulLO is suggested to function in hydrogen peroxide generation in nectar but not in plant ascorbate biosynthesis.

Proteomics and gene expression analysis of Nicotiana tabacum nectar proteins

Project description:Floral nectar proteins (nectarins) are mainly enzymes and play important roles in inhibiting microbial growth in nectar and tailoring nectar chemistry before or after secretory. Nectar proteomes are usually small, but only very few plant species have had their nectar proteomes thoroughly investigated. Nectarins from Nicotiana tabacum (NT) were separated using two-dimensional gel electrophoresis, and then analyzed using mass spectrometry. Glycoproteins were isolated from raw NT nectar, separated by SDS-PAGE, and identified by mass spectrometry. All eight identified nectarins and four invertase genes’ expression were analysed by qPCR. Sugars composition, total sugar concentration, protein content, polyphenol content and hydrogen peroxide content were compared at different time intervals in extracted nectar and nectar in situ after secretion. Totally, eight nectarins were detected in NT nectar in which only two are glycoproteins, beta-xylosidase and a protein with unknown function. All of the eight nectarin genes expression was not nectary-specific and not synchronous along with the nectary development. After secretion, NT nectar in flower tube changed from sucrose–rich to hexose-rich type even though no free invertase or its activity was detected in NT nectar. No sugar composition changes observed in extracted nectar after incubating at 30 ℃ up to 48 hours in plastic tubes. Our results indicate that nectar post-secretory changes could be a complex process and tissue closely contact with nectar might function in it.

ESCRTIII-associated protein AtALIX mediates high affinity phosphate transporter trafficking to maintain phosphate homeostasis in Arabidopsis

Project description:Prior to the release of their cargoes into the vacuolar lumen, sorting endosomes mature into multivesicular bodies (MVB) through the action of ENDOSOMAL COMPLEX REQUIRED FOR TRANSPORT (ESCRT) protein complexes. MVB-mediated sorting of high affinity phosphate transporters (PHT1) to the vacuole limits their plasma membrane levels under phosphate sufficient conditions, a process that allows plants to maintain phosphate homeostasis. We describe here AtALIX, a cytosolic protein that associates to MVB by interacting with ESCRT-III subunit SNF7 and enables PHT1;1 trafficking to the vacuole. Thus, we show that the partial loss of function mutant Atalix-1 displays reduced vacuolar degradation of PHT1;1. AtALIX versions containing the Atalix-1 mutation showed reduced interaction with SNF7, providing a simple molecular explanation for impaired cargo trafficking in Atalix-1 mutants. Indeed, Atalix-1 mutation also hampered vacuolar sorting of brassinosteroid receptor BRI1. In line with a presumed broad target spectrum, Atalix-1 mutation is pleiotropic, provoking reduced plant growth, late flowering and altered vacuole morphogenesis, whereas null mutants are lethal, indicating that AtALIX controls diverse processes in plants being essential for their life.

Differential gene expression in Leishmania [SPP.] promastigotes. Leishmania chagasi

Project description:Leishmania chagasi is the causative agent of zoonotic visceral leishmaniasis in Brazil, being domestic and stray dogs the main reservoirs. The development of the parasite involves two stages. The promastigote is extracellular and develops within the sand fly gut. The amastigote survives inside the harsh environment of the phagolysosome of mammalian host phagocytes, where pH is acidic, temperature higher than in the sand fly vector and hydrolytic enzymes act. In addition, the host phagocyte displays the nitric oxide defense mechanism against the amastigote. Promastigotes are also able to withstand NO even when they develop within the sand fly gut. This can be explained with the pre-adaptative hypothesis, which has been supported by us and others elsewhere and consists of preparation of promastigotes in advance for development towards the amastigote stage. For this reason, the comparison of NO-resistant and sensitive promastigotes is valuable. The two-dimension electrophoresis-mass spectrometry (2DE-MS/MS) approach has been performed to study differential protein abundance comparing L. chagasi NO-sensitive and resistant promastigotes. This analysis has revealed differential expression of genes directly and indirectly involved in NO-resistance, highlighting up-regulation of the glucose-6-phosphate dehydrogenase (G6PD) in NO-resistant promastigotes and down-regulation of the glutathione peroxidase (GPX) and the arginase (ARG) in NO-sensitive ones. These data are a starting point in the search of vaccine candidates and/or drug targets.
