Project description:The DBA/1 mouse strain produces spontaneous arthritis (SpAD) characterized by an inflammatory process and changes in the joint structures that resemble human spondyloarthritis. Normally, it is the hind paws that are analyzed in this model and information on the alterations in the knee is very scarce. Transcriptome analysis was performed using DNA microarrays of knees of DBA/1 strain mice with spontaneous arthritis (SpAD) and BALB/c strain mice as a control.

Early gene expression differences in inbred mice predict susceptibility to spontaneous pulmonary adenomas

Project description:We examined if gene expression profiles differed between healthy (tumor-free) mice of strains that are known to have different pulmonary adenoma susceptibility (severity and frequency). Three females of 11 week old mice of 9 (129S1/SvlmJ, A/J, BALB/cJ, C3H/HeJ, C57BL/6J, C57L/J, DBA/2J, KK/HlJ, and MRL/MpJ) strains were selected for RNA extraction and hybridization on GeneChip® Mouse Gene 1.0 ST Arrays (Affymetrix, Santa Clara, CA). Each strain was compared to all other strains (36 unique comparisons total) to identify the most common differentially expressed probe sets among the population. The most common probesets were then compared between strain groups (increased vs. decreased spontaneous pulmonary adenoma frequency and severity).

Transcription profiling of mouse F2 intercross mice generated from arthritis-prone BALB/c and arthritis-resistant DBA/1 mice deficient for interleukin 1 receptor antagonist (Il1rn)

Project description:Rheumatoid arthritis (RA) is a heterogeneous disease with clinical and biological polymorphisms. However, little is known about baseline molecular variations among individual RA patients. The purpose of this study was to address this issue using F2 intercross mice generated from arthritis-prone BALB/c and arthritis-resistant DBA/1 mice deficient for interleukin 1 receptor antagonist (Il1rn). Two distinct subpopulations of arthritic mice were identified in the 38 mice studied. One subgroup of diseased mice was characterized by myeloid cell dominant inflammation, whereas the other was mainly associated with increased anti-apoptotic activities of inflammatory cells. Experiment Overall Design: Thirteen F2 mice, 18 with arthritis and 18 without, were included in this study. Total RNAs were extracted from individual spleens for Affymmetrix mouse genome array analysis. Normalized data were hierarchically clustered to identify subpopulations of arthritic mice.

Whole-genome RNA-deep sequencing of pancreatic islets of congenic Nidd/DBA mice

Project description:Here, we describe the QTL Nidd/DBA, a diabetogenic allele which was contributed by DBA and enhanced hyperglycemia and β-cell loss. In order to re-address the issue of which variants on chromosome 4 are responsible for this trait, Nidd/DBA recombinant congenic lines were generated on an NZO background. Gene expression profiles of pancreatic islets were generated from congenic mice carrying the initial 13.6 Mbp fragment (RCS-I) of the Nidd/DBA locus. Six genes within the critical region were differentially expressed in pancreatic islets of homozygous Nidd/DBAD/D mice compared to homozygous Nidd/DBAN/N controls.
