Novel Epigenetic Mechanisms of Endogenous CSE/H₂S in Smooth Muscle Cells for Abdominal Aortic Aneurysm

Project description:Background: The incidence of abdominal aortic aneurysm is increasing year by year. Its rupture is associated with a high mortality rate, and recurrence is extremely likely even after treatment. The gasotransmitter hydrogen sulfide (H₂S) and its key generating enzyme, cystathionine -γ -lyase (CSE), possess anti -inflammatory, antioxidant, and anti -apoptotic properties, and have a cardiovascular protective effect. However, the role of CSE in AAA development and progression remains elusive. Methods: Using smooth muscle cell-specific CSE-knockout mice as models and angiotensin II, β-aminopropionitrile as inducers, it was found that CSE knockout in smooth muscle cells increased AAA incidence and mortality. Experimental results confirmed smooth muscle endogenous CSE/H₂S is involved in AAA development. However, its molecular mechanism remains a key scientific issue urgently needing solution. Thus, whole-genome RNA sequencing analysis was carried out on primary aortic smooth muscle cells isolated from Ang II-stimulated C57BL/6J mice (CSE WT) and global CSE-knockout mice (CSE KO) to identify potential key downstream genes regulated by CSE. Results: The RNA - seq data highlighted the crucial role of CSE in the regulation of extracellular matrix and the PRC complex, which has implications for the development of AAA.

Project description:In this study, ChIP - seq was employed to investigate the impact of CSE on epigenetic modifications in vascular smooth muscle cells (VSMCs) and to uncover its influence on genome - wide DNA binding. We conducted ChIP - seq experiments on H3K9me3, H3K9ac, and H3K4me3 using primary smooth muscle cells from CSE knockout mice and wild - type (WT) mice after 24 - hour stimulation with Ang II. Our finding demonstrated that endogenous CSE/H₂S in smooth muscle regulates the expression of extracellular matrix through epigenetic modifications and participates in the occurrence and development of abdominal aortic aneurysm (AAA).

Project description:An abdominal aortic aneurysm (AAA) is a pathological dilatation of the aortic wall and it is a life-threatening disease due to the risk of rupture. Currently, surgical intervention remains the only definitive treatment recommended for large or rapidly expanding aneurysms. No pharmacological therapy is currently available to prevent AAA progression, as no drug has been proven effective. Diabetes is a major risk factor for cardiovascular disease in general. However, many studies over the last decade show that type 2 diabetes patients have a lower incidence of AAA and a slower aneurysm growth rate, which has been correlated with metformin use. We performed a proteomics analysis on resected aortic tissue from controls, non-diabetic (ND) and diabetic (D) AAA patients.

Mitochondrial NAD+ deficiency in vascular smooth muscle impairs collagen III turnover to trigger thoracic and abdominal aortic aneurysm

Project description:Thoracic and abdominal aortic aneurysm poses a substantial mortality risk in adults, yet many of its underlying factors remain unidentified. Here, we identify mitochondrial nicotinamide adenine dinucleotide (NAD)⁺ deficiency as a causal factor for the development of aortic aneurysm. Multiomics analysis of 150 surgical aortic specimens indicated impaired NAD+ salvage and mitochondrial transport in human thoracic aortic aneurysm, with expression of the NAD+ transporter SLC25A51 inversely correlating with disease severity and postoperative progression. Genome-wide gene-based association analysis further linked low SLC25A51 expression to risk of aortic aneurysm and dissection. In mouse models, smooth muscle-specific knockout of Nampt, Nmnat1, Nmnat3, Slc25a51, Nadk2 and Aldh18a1, genes involved in NAD+ salvage and transport, induced aortic aneurysm, with Slc25a51 deletion producing the most severe effects. Using these models, we suggest a mechanism that may explain the disease pathogenesis: the production of type III procollagen during aortic medial matrix turnover imposes a high demand for proline, an essential amino acid component of collagen. Deficiency in the mitochondrial NAD⁺ pool, regulated by NAD⁺ salvage and transport, hinders proline biosynthesis in mitochondria, contributing to thoracic and abdominal aortic aneurysm.
