Project description:Dietary components influence microbial composition in the digestive tract. Although often viewed as energy sources, dietary components are likely to shape microbial determinants of intestinal colonization beyond metabolism. Here, we report that a dietary long-chain fatty acid enhances the yeast Candida albicans colonization of the murine gut partly by eliciting modifications to the fungal cell surface. Mice fed an oleic acid-rich diet were readily colonized by C. albicans and exhibited higher fungal load in feces compared to rodents fed an isocaloric control diet. Surprisingly, β-oxidation, a catabolic process to break down fatty acids for energy production, was dispensable for C. albicans to colonize the high-oleic acid diet-fed mice. RNA-Seq was employed to identify oleic acid-induced transcriptome changes under anaerobic conditions. We identified SOK1 as an oleic acid-induced kinase that dictates cell wall mannan exposure and binding to intestinal mucin under anaerobic conditions. Furthermore, oleic acid induced the expression of several C. albicans transcription factors that positively regulate intestinal colonization via remodeling of the fungal cell surface.

RNA-seq of Candida albicans Hda1 complex mutants (hda1∆∆, hda2∆∆ and hda3∆∆) and wildtype SN152 in yeast and hyphae inducing conditions

Project description:Candida albicans is a normally commensal yeast which becomes pathogenic to human hosts under some conditions and specifically in immune compromised individuals. This contributes to a global health crisis of fungal disease. This yeast can morphologically adapt to the environment by reversibly developing hyphae which allows for traversing of tissue. Histone deacetylase 1 (Hda1) is involved in this transition as it is established that the deletion of the associated gene stunts this morphological switch. In Saccharomyces cerevisiae this protein complexes with two similarly named proteins: Hda2 and Hda3. Here we evaluate Candida albicans strains with mutations in the genes that code for these proteins. The strains are grown in yeast (YPD 30 °C) and hyphae (RPMI 37 °C) -inducing conditions for 90 minutes prior to RNA extraction. After alignment by HISAT2, the homozygous mutants are then individually compared to wildtype (matching growth conditions) using DESeq2.

Project description:The effect of the deletion of the hyphae-associated toxon of Candida albicans, camdidalysin, on yeast and hyphae tramscriptomes was tested with different mutants. Tested strains include the wild type (SC5314), deletion mutants of candidalyin and of ECE1 (the gene encoding the polyprotein that includes candidalysin) and an ECE1 revertant strain. Each were tested in yeast- and in hypae-supporting growth conditions.

Expression profiling of Candida albicans and Candida dubliniensis in reconstituted human oral epithelium 90 min p.i.

Project description:Candida albicans and Candida dubliniensis are closely related species displaying differences in virulence and genome content, therefore providing potential opportunities to identify novel C. albicans virulence genes. C. albicans gene arrays were used for comparative analysis of global gene expression in the two species in reconstituted human oral epithelium (RHE). C. albicans (SC5314) showed upregulation of hypha-specific and virulence genes within 30 min postinoculation, coinciding with rapid induction of filamentation and increased RHE damage. C. dubliniensis (CD36) showed no detectable upregulation of hypha-specific genes, grew as yeast, and caused limited RHE damage. Several genes absent or highly divergent in C. dubliniensis were upregulated in C. albicans. One such gene, SFL2 (orf19.3969), encoding a putative heat shock factor, was deleted in C. albicans. ΔΔsfl2 cells failed to filament under a range of hypha-inducing conditions and exhibited greatly reduced RHE damage, reversed by reintroduction of SFL2 into the ΔΔsfl2 strain. Moreover, SFL2 overexpression in C. albicans triggered hyphal morphogenesis. Although SFL2 deletion had no apparent effect on host survival in the murine model of systemic infection, ΔΔsfl2 strain-infected kidney tissues contained only yeast cells. These results suggest a role for SFL2 in morphogenesis and an indirect role in C. albicans pathogenesis in epithelial tissues.
