Project description:Drought stress is a major environmental factor limiting crop productivity. Arbuscular mycorrhizal fungi (AMF), as beneficial soil microbes, can improve plant growth and stress resilience; however, the effectiveness of this symbiosis is often influenced by the host plant's genetic background. In this study, we investigated the interaction between AM symbiosis and drought tolerance in two foxtail millet (Setaria italica) genotypes with contrasting drought responses: the drought-tolerant ISE42 and the drought-sensitive TT8. Following a 14-day drought treatment, both genotypes exhibited wilting, but only AMF-colonized ISE42 plants recovered upon rewatering. Transcriptomic analysis revealed that AM symbiosis significantly enhanced the expression of genes involved in nitrogen transport, assimilation, lignin metabolism, and cellulose biosynthesis in ISE42, but not in TT8. These molecular changes suggest improved nutrient uptake and cell wall reinforcement as key mechanisms underlying enhanced drought tolerance. Additionally, stress hormone signaling pathways were downregulated in colonized ISE42 roots, indicating possible alleviation of drought-induced stress through AM symbiosis. Our results demonstrate genotype-specific effects of AMF on drought tolerance and highlight the importance of considering host genetic variation in the application of AMF for crop improvement.

We then performed gene expression profiling analysis using data obtained from RNA-seq of 3 different landraces (Hanevalval, TT8, ICE36) after 6-week mock or arbuscular mycorrhizal fungi treatments.

Variation of growth and transcriptome responses to arbuscular mycorrhizal symbiosis in different foxtail millet lines.

Project description:BackgroundArbuscular mycorrhizal fungi (AMF) have been applied to promote the growth of different crop species, but knowledge about the impacts of symbiosis on foxtail millet at the physiological and molecular levels have remained limited. In this study, we compared the mycorrhization phenotypes of one cultivar and three different landraces and performed a comprehensive transcriptomic analysis to assess the effects of genetic variation on the responses to symbiosis.ResultsOur results showed that colonization by AMF did not enhance biomass accumulation but significantly increased grain production only in three lines. More than 2,000 genes were affected by AMF colonization in all lines. Most AM symbiosis-conserved genes were induced, but the induction levels varied between lines. Gene Ontology (GO) analysis showed that Biological Function terms related to nitrogen transport and assimilation were only enriched in TT8. Similarly, two of phosphate starvation-induced phosphate transporters were only simultaneously downregulated in TT8. In the other two lines, the enrichment of GO terms associated with cell wall reorganization and lignification was observed, though the effects were different.ConclusionThis study reveals the impacts of genetic variation of millet lines on the responses to AM symbiosis and provides information regarding AMF application for millet production.

Moreover, through transcriptome sequencing and Data-independent acquisition (DIA) quantitative proteomics determination of sinrx1 mutants and wild types (WT) at the seedling stage under drought and control conditions, the molecular mechanism of SiNRX1 regulating drought resistance was preliminarily analyzed.
