Project description:Transgenic (Tg) mice over-expressing TGFb1 in a lung-specific manner display airway inflammation and fibrosis, phenotypes relevant to human COPD and asthma. This model system provides a platform for identifying and validating genes/pathways involved in these disease pathogeneses. The TGFb1-Tg mice lung profiling data was analyzed by identifying genes that were up- and down-regulated at selected p value and fold change in the lung of TGFb1-Tg mice compared to the corresponding wild type controls.

Project description:This project aims to compare gene expression in transgenic mice that overexpress TGFB1 to wild type mice. Transgenic mice have a fibrotic response in the atria, but not the ventricle. This is a 2 color design; dye swaps and a loop design were incorporated. Each group has N=4, with the exception being WT atria which has N=3 and was hybridied twice, the groups are Wt atria, WT ventricle, Transgenic atria, Transgenic ventricle. Comparisons of interest : Tg atria vs WT atria, Tg ventricle vs WT ventricle, WT atria vs.dWT ventricle, Tg atria vs Tg ventricle
Project description:To investigate the differences in microRNA expression profiles between fibrotic and normal livers, we performed microRNA microarrays for total RNA extracts isolated from mouse livers treated with carbontetrachloride (CCl4) or corn-oil for 10 weeks (n=3/group). MicroRNAs were considered to have significant differences in expression level when the expression difference showed more than two-fold change between the experimental and control groups at p<0.05. We found that 12 miRNAs were differentially expressed in CCl4-induced fibrotic liver. To induce chronic liver fibrosis, seven-week-old mice received 0.6 ml/kg body weight of carbon-tetrachloride (CCl4) dissolved in corn-oil by intraperitoneal (i.p.) injection, twice a week for 10 weeks (n=3). As a control, same number of mice was injected with equal volume of corn-oil for 10 weeks.
