Cytoplasmic lysates were separated into monosome, light-polysome, and heavy-polysome fractions, ERCC spike-ins were added after fractionation and before RNA extraction, and fraction RNA was sequenced to quantify transcript ribosome-load state. The processed matrix reports control fraction distributions, fraction-enrichment classifications, polyEN, and mean ribosome load (MRL).
