Project description:The gene encoding elongation factor G, fusA1, is frequently mutated in clinical isolates of Pseudomonas aeruginosa from patients with cystic fibrosis. Recent work has shown that fusA1 mutants often display elevated aminoglycoside resistance due to increased expression of the aminoglycoside efflux pump, MexXY. We isolated a spontaneous gentamicin-resistant fusA1 mutant (FusA1-P443L) in which mexXY expression was increased. We compared the proteome of this fusA1 mutant (EMC1) with the P. aeruginosa PAO1-derived progenitor strain (EMC0) and complemented mutant strain expressing the wild-type fusA1 gene in trans (EMC1*).

Project description:We present here a transcriptional regulator, PA3898, which controls biofilm formation and multidrug efflux-pumps in P. aeruginosa. A mutant of this regulator significantly reduced the ability of P. aeruginosa to produce biofilm in vitro, and affected its in vivo fitness and pathogenesis in Drosophila melanogaster and BALB/c mouse lung infection models. Transcriptome analysis revealed that PA3898 modulates essential virulence genes/pathways, including multidrug efflux-pumps and phenazine biosynthesis.

Expression profiles of Pseudomonas aeruginosa TBCF10839 wild type and nadk1 mutant under oxidative stress conditions

Project description:Phagocytosis and killing of the opportunistic pathogen Pseudomonas aeruginosa by polymorphonuclear neutrophils (PMNs) is the major antipseudomonal host defense of vertebrates. By screening a transposon library of the clinical P. aeruginosa isolate TBCF10839 that can grow and replicate in PMNs, a mutant was identified that was most strongly sensitized to killing by PMNs. The inactivated gene PA1572 termed nadK1 was found to encode an ATP:NAD kinase. The transcriptomes of the TBCF10839 wild type and nadK1 mutant were investigated in the presence of PMNs or H2O2. Exposure to H2O2 led to diametrical mRNA expression profiles. H2O2-degrading enzymes were upregulated by wild type, but not by nadK1 mutant. This data demonstrates that NadK1 is crucial for the response of P. aeruginosa to reactive oxygen species.
