Project description:Streptococcus suis is an important zoonosis pathogen that causes significant economic losses worldwide characterized by meningitis, septicaemia, arthritis, bronchopneumonia endocarditis. Streptcoccus suis 2 strain SC19 was isolated in Sichuan province in China, during the outbreak in 2005. Septicemia is most popular symptoms for SC19 infection, and mortality is high. We used human acute monocytic leukemia cell line (THP-1) infected SC19 to analysis the pathomechanism of septicemia in SS2 infection.

During the infection the parasite releases a number of antigens (called Excretory Secretory Products - ESP) facilitating the parasites survival. A number of ESP components have immunomodulatory properties. To determine if ES from various F. hepatica isolates have different immunomodulatory abilities LPS activated “BOMA” cells were stimulated with ES of laboratory Weybridge (FhWey-ES) and wild (FhWild-ES) Fasciola hepatica isolates.

Project description:Leptospirosis is a re-emerging zoonosis, a globally important infectious disease, caused by an infection with the genus Leptospira. Leptospirosis is associated with acute kidney injury and progress to CKD due to sustained tubulointerstitial inflammation. Macrophages play a critical role in controlling the bacterial burden and tissue inflammation during the spirochete infections. To understand the molecular mechanisms of leptospia-induced macrophage activation and its role in the inflammatory process, we performed the transcriptome profiles of murine bone marrow-derived macrophages infected with pathogenic and non-pathogenuc Leptospira spp.at a multiplicity of infection of 100 for 2 and 24 hrs, respectively.
Project description:Fasciola hepatica is the agent of fasciolosis, a foodborne zoonosis that affects livestock production and human health. Although flukicidal drugs are available, re-infection and expanding resistance to triclabendazole demand new control strategies. Understanding the molecular mechanisms underlying the complex interaction with the mammalian host could provide relevant clues, aiding the search for novel targets in diagnosis and control of fasciolosis.
Project description:A novel henipavirus (HNV) named Langya virus (LayV) was isolated in human patients in China in August 2022. It is closely related to Mòjiāng virus (MojV) and represents the first instance of HNV zoonosis to humans outside of Nipah virus (NiV) and Hendra viruses (HeV). Within this work, mass spectrometry glycoproteomic analysis revealed that although the LayV F protein has reduced glycosylation compared to its NiV and HeV counterparts, a unique glycan was identified positioned at the DIII apex, shielding a previously identified site of vulnerability in NiV F.

Human acute monocytic leukemia cell line (THP-1) cells were stimulated with Streptcoccus suis 2 (SS2) strain SC19. We added SS2 to THP-1 cells at a MOI of 1:1 (bacteria/cells). Uninfected control cells were incubated with PBS only. After 3 hours incubation, cells were collected for RNA extraction and hybridization on Affymetrix microarrays. A total of 4 samples were challenged, and 4 samples were used as controls. 4 microarrays were used in this experiment.
