Project description:Despite recent advances, molecular events and mechanisms involved in long-lasting memory formation are unclear. Here, we report a critical role for nucleosome remodeling factor BAF53b in mice. We observed an increase of BAF53b expression beyond 24 h after fear conditioning in the lateral amygdala, which was crucial for memory retention at remote time. RNA-seq analyses and behavioral rescue experiments identified fibroblast growth factor-1 (FGF1) as a candidate downstream effector.

Optogenetic Stimulation of Prelimbic Pyramidal Neurons Maintains Fear Memories and Modulates Amygdala Pyramidal Neuron Transcriptome

Project description:The suppression of fear memory in the absence of danger (fear extinction) requires coordinated neural activity within the amygdala and medial prefrontal (prelimbic and infralimbic) cortex. Any behavior has a transcriptomic signature that is modified by environmental experiences, and specific genes are involved in functional plasticity and synaptic wiring during fear memory and extinction. In the present study, we investigated the effects of optogenetic manipulations of prelimbic pyramidal neurons on amygdala gene expression to analyze the specific transcriptional pathways involved in fear extinction. To this aim, transgenic mice (Thy1-COP4) having cortical and amygdala pyramidal neurons optogenetically excitable were (or not) fear-conditioned. During the extinction phase, the mice received optogenetic (or sham) stimulations to maintain the activation of the prelimbic pyramidal neurons and impair fear extinction. At the end of behavioral testing, electrophysiological (Excitatory Post-Synaptic Currents) and morphological (spinogenesis) correlates were evaluated in the pyramidal neurons of prelimbic cortex. Furthermore, transcriptomic cell-specific RNA-analyses (differential gene expression profiling and functional enrichment analyses) were performed in amygdala pyramidal neurons. Results demonstrate that pyramidal neurons of prelimbic cortex are involved in modulation of the fear responses during extinction phase and their optogenetic stimulation in fear-conditioned mice results in strong modifications of the amygdala transcriptome. Understanding the transcriptomic architecture of fear extinction may facilitate the comprehension of fear-related disorders.

Analysis of amygdala astrocytes as a function of stress and fear

Project description:Neuroimmune interactions—signals transmitted between immune and brain cells—regulate many aspects of tissue physiology including responses to psychological stress, which can predispose individuals to develop neuropsychiatric diseases. Still, the interactions between hematopoietic and brain-resident cells that influence complex behaviors are poorly understood. Here, we used a combination of genomic and behavioral screens to demonstrate that astrocytes in the amygdala limit stress-induced fear behavior through EGFR. Mechanistically, amygdala astrocyte EGFR expression inhibits a stress-induced pro-inflammatory signal transduction cascade that facilitates neuro-glial cross-talk and stress-induced fear behavior through the orphan nuclear receptor NR2F2 in amygdala neurons. We uncovered that, in turn, decreased EGFR signaling and fear behavior were associated with meningeal monocyte recruitment during chronic stress. This set of neuroimmune interactions was therapeutically targetable by psychedelic administration, which reversed monocyte accumulation in the brain meninges along with fear behavior. Together with validation in clinical samples, these data suggest that psychedelics can be used to target neuroimmune interactions relevant to neuropsychiatric disorders and potentially other inflammatory diseases.
