Project description:We generated adeno-associated Cre viral vectors with improved astrocyte-specific expression by including a cassette of miRNA targeting sequences (4x6T) to decrease off-target expression. We delivered these viruses in Ribotag mice using the PHP.eB serotype, either intracortically or systemically. We compared these samples with mice that received intracortical AAV2/5::GfaABC1D-Cre and transgenic Aldh1l1-CreERT2/Ribotag mice. We sequenced whole-tissue RNA as well as ribosomally-loaded astrocyte-enriched mRNA to assess cell-type specificity of viruses as well as virus-related transcriptomic changes.

Neuronal conversion of pre-labeled astrocyte using adeno-asociated viruses in vivo

Project description:Neuronal reprogramming using adeno-associated viruses (AAVs) carrying a GFAP mini-promoter to drive neurogenic transcription factor expression in astrocytes in vivo were refuted due to off-target neuronal expression. Here, we show successful neuronal conversion of pre-labeled astrocytes using AAV-GFAP to express phospho-deficient Ascl1SA6, and demonstrate that miR124 target sites reduce off-target neuronal expression. Finally, single cell transcriptomic and pseudotime trajectory analyses confirmed Ascl1SA6-driven astrocyte-to-neuron conversion, and more limited oligodendrocyte conversion events

Project description:We generated a stable healthy and diseased (with ALS-linked FUS-R521H mutation) iPSC line containing the coding sequence of SOX9 under a Tet-on promotor in the safe harbor AAVS1 locus via recombinase-mediated cassette exchange. This allows the induction of SOX9 expression after doxycycline addition and generates in this way PSC-derived astrocytes. RNASeq is performed on these healthy and diseased iSOX9-astrocytes at an early and late timepoint. In addition, we included commercially available PSC-derived astrocytes (called "iCell") and fetal human primary astrocytes (called "fHA").
