Identifying target proteins of capsazepine using DARTS technology

ABSTRACT: This study employs DARTS (Drug Affinity Responsive Target Stability) to investigate capsazepine's targets. The technique detects drug-induced proteolytic resistance in target proteins, coupled with mass spectrometry to capture drug-protein complexes. Experiments focus on identifying novel targets beyond TRPV1 (e.g., G protein-coupled receptors) to elucidate its analgesic mechanisms and polypharmacological effects. Key findings may advance pharmacological optimization of analgesics and expand therapeutic applications.

Eugenol modulates the NOD1- NF-κB signaling pathway via targeting NF-κB protein in triple breast cancer cells

Project description:Background: The most aggressive subtype of breast cancer, triple-negative breast cancer (TNBC), has a worse prognosis and a higher probability of relapse since there is a narrow range of treatment options. Identifying and testing potential therapeutic targets for the treatment of TNBC is of high priority. Methods: Using a transcriptional signature of triple-negative breast cancer collected from Gene Expression Omnibus (GEO), CMap was utilized to reposition compounds for the treatment of TNBC. CCK8 and colony formation experiments were performed to detect the effect of the candidate drug on the proliferation of TNBC cells, while flow cytometry was used to detect cell apoptosis. Meanwhile, transwell assay was implemented to detect cell metastasis change caused by the candidate drug. Moreover, the proteomic approach was presently ongoing to evaluate the underlying mechanism of the candidate drug in TNBC. Furthermore, drug affinity responsive target stability (DARTS) coupled with LC-MS/MS was carried out to explore the potential drug target candidate in TNBC cells. Results: We found that the most widely used medication, eugenol, reduced the growth and metastasis of TNBC cells. According to the underlying mechanism revealed by proteomics, eugenol could inhibit TNBC cell proliferation via the NOD1-NF-κB signaling pathway. DARTS experiment further revealed that eugenol may bind to NF-κB in TNBC cells. Concludes: Our findings pointed out that eugenol was a potential candidate drug for the treatment of TNBC.

Nitidine chloride inhibits mTORC1 signaling through ATF4-mediated Sestrin2 induction and targets IGF2R for autophagic degradation

Project description:Nitidine chloride (NC), a natural phytochemical alkaloid derived from Zanthoxylum nitidum (Roxb.) DC, exhibits multiple bioactivities in cancer, infection disease, and other therapeutic areas. However, the primary targets of NC and the mechanism of action (MOA) have not been defined explicitly. Here, we report that NC inhibit mTORC1 activity by targeting amino acid sensing signaling through activating transcription factor 4 (ATF4)-mediated Sestrin2 induction, and it efficiently induces autophagy in cells and tumor xenograft. NC displays potent cytotoxicity against various cancer cells and inhibits xenograft tumor growth of B16 melanoma. Furthermore, we identified insulin-like growth factor 2 receptor (IGF2R) as a direct target of NC by drug affinity responsive target stability (DARTS) technique. NC may functional as a IGF2R antagonist to inhibit growth factor-mediated mTORC1 activation. This study suggested that NC targets mTORC1 signaling through nutrients sensing and growth factor receptor binding, providing mechanistic insights into the MOA of NC.

Project description:Cannabidiol (CBD) showed antiproliferative activity and induction of apoptosis in chronic myelogenous leukaemia cancer cells (K562 cells). A consolidated DARTS (Drug Affinity Response Target Stability) and t-LIP-MRM (targeted- Limited Proteolysis-Multiple Reaction Monitoring Mass spectrometry) platform was applied to unveil the interactome of CBD and to shed light on its mechanism of action. Both approaches point to the identification of DNA topoisomerase 2-alpha (TOP2A) as a major CBD target in K562 cells.

NSUN2 Facilitates Dicer Cleavage of DNA Damage-Associated R-Loops to Promote Repair [ONT direct RNA sequencing]

Project description:DNA integrity is constantly challenged by both endogenous and exogenous damaging agents, resulting in various forms of damage. Failure to repair DNA accurately leads to genomic instability, a hallmark of cancer. Distinct pathways exist to repair different types of DNA damage. Double-strand breaks (DSBs) represent particularly severe form of damage, due to physical separation of DNA strands. Repair of DSBs requires activity of RNA Polymerase II (RNAPII) and generation of Damage-associated transcripts (DARTs). Here we show that the RNA m5C-methyltransferase NSUN2 localizes to DSBs in transcription-dependent manner, where it binds to and methylates DARTs. Depletion of NSUN2 results in an accumulation of nascent DARTs around DSBs, specifically of the de novo primary DARTs. Furthermore, we detected an RNA-dependent interaction between NSUN2 and DICER, which was stimulated by DNA damage. NSUN2 activity promoted Dicer cleavage of DARTs associated R-loops, which is required for efficient DNA repair. We report a previously unrecognized role of the RNA m5C-methyltransferase NSUN2 within RNA-dependent DNA damage response, highlighting its function as a DICER chaperone for the clearance of non-canonical substrates such as DARTs, thereby contributing to genomic integrity.
