Multi-omics toxicity profiling of engineered nanomaterials

Project description:This study aims to implement methods for toxicological profiling of engineered nanomaterials using toxicogenomics, proteomics, and metabolomics along with computational analyses. For all three omics layers the human cell lines A549 (lung epithelial cells) and THP1 (monocytes) were separately exposed to the nanomaterials TiO2 NM104, MWNCT NM401, and Ag NM300k. Proteomics and metabolomics samples have been performed as biological triplicates and were taken after 0, 6, 12, 24, and 48 hours. To assess ecotoxic effects C. elegans worms were grown in soil treated with NM300k. Ecotox samples were taken only at 0 and 24 hours. Integrating all three omics layers will enable the identification of (novel) ENM specific modes of action (MoA).

Project description:In this study we exposed three human lung derived cell lines to sublethal dosages of nanomaterials for a limited amount of time. For the first time, we assessed the simultaneous effects of nanomaterials exposure on three distinct molecular layers along with their interactions in the determination of the MOA of 10 carbon based nanomaterials. By performing an integrative analysis we provide here a complete picture of the interaction between regulatory factors (DNA methylation and miRNAs) and mRNA deregulation subsequent to exposing different cellular systems to different nanomaterials.

Multi-hierarchical profiling the structure-activity relationships of engineered nanomaterials at nano-bio interfaces

Project description:Increasing concerns over the possible risks of nanotechnology necessitates breakthroughs in structure-activity relationship (SAR) analyses of engineered nanomaterials (ENMs) at nano-bio interfaces. However, current nano-SARs are often based on univariate assessments and fail to provide tiered views on ENM-induced bio-effects. Here we report a multi-hierarchical nano-SAR assessment for a representative ENM, Fe2O3, by metabolomics and proteomics analyses. The established nano-SAR profile allows the visualizing of the contributions of seven basic properties of Fe2O3 to its diverse bio-effects. For instance, although surface reactivity is responsible for Fe2O3-induced cell migration, the inflammatory effects of Fe2O3 are determined by aspect ratio (nanorods) or surface reactivity (nanoplates). These nano-SARs are examined in THP-1 cells and animal lungs, which allow us to decipher the detailed mechanisms including NLRP3 inflammasome pathway and monocyte chemoattractant protein-1-dependent signaling. This study provides more insights for nano-SARs, and may facilitate the tailored design of ENMs to render them desired bio-effects.

Reproducible label-free quantitative proteomics profiling cellular responses to engineered nanomaterials

Project description:Proteomic data from engineered nanomaterial- treated THP-1 cells. Time points at 12 hours; 4 biological replicates for each treatment. Human THP-1 cells were either mock-treated (control) or treated with engineered nanomaterials (ENMs). Four biological replicates were used for each sample group (Control, or ENM treatment at either the low or high dosage). In the block design scheme for LC-MS analysis, samples treated with two different ENMs were assign into an analytical block (2 ENM X 2 dosages/ENM X 4 replicates = 16 samples). Four control samples were also included in each block, yielding 20 samples per block. Blocks of samples were divided into two processing batches. Samples in each batch were processed simultaneously from cell culture, to treatment with ENMs, and finally to proteomic sample preparation for LC-MS analysis (including trypsin digestion).
