ABSTRACT: This dataset contains normalized protein expression (NPX) values from Olink proteomic profiling of serum from patients with major depressive disorder, atopic dermatitis, psoriasis, and healthy controls, using the proximity extension assay technology across four panels: Target 96 Inflammation, Cardiovascular II, Cardiovascular III, and Neurology.

Major depressive disorder (MDD) is a prevalent neuropsychiatric disorder associated with significant morbidity and mortality. Increasing evidence suggests that a subset of MDD patients exhibit a dysregulated immune system. Targeted Plasma Proteomics Reveals a Central Role of Upregulated TNFRSF Proteins in HIV-associated Stroke: A Cross-sectional Discovery and Longitudinal Validation Study

Project description:This study investigated circulating proteomic signatures associated with stroke in people with HIV (PWH) using a nested case–control design. Plasma samples were collected from 135 HIV-positive individuals with stroke (HIV+/Stroke+), 135 HIV-positive stroke-free controls (HIV+/Stroke−), and 135 HIV-negative stroke-free controls (HIV−/Stroke−) from the CHART cohort (Taizhou, China). A longitudinal sub-cohort of 37 HIV+/Stroke+ individuals with paired pre- and post-stroke samples was additionally included for temporal validation. All plasma samples were profiled using the Olink Target 96 Cardiovascular II and Cardiovascular III panels simultaneously, quantifying 184 non-overlapping protein biomarkers. The study aimed to identify stroke-associated proteomic signatures, characterise underlying biological pathways and protein interaction networks, and validate key proteins longitudinally.

Project description:Here, we present an in-depth proteomic profiling of pre-diagnostic plasma samples from 88 individuals with suspected lung cancer, including 44 patients diagnosed with stage III-IV lung adenocarcinoma (LUAD) and 44 non-cancer controls. Proteomic analysis was performed using Olink Explore 3072, which comprises four distinct 384-plex panels targeting inflammation, oncology, cardiometabolic, and neurology proteins.

Longitudinal Olink affinity proteomics profiling of cytokine release syndrome in myeloma patients receiving BCMA-directed CAR-T therapy

Project description:This submission contains de-identified Olink NPX affinity proteomics data from longitudinal peripheral blood samples collected from patients with relapsed/refractory multiple myeloma treated with BCMA-directed CAR-T therapy (ide-cel or cilta-cel) in a prospective single-center pilot study. Samples were analyzed using the Olink Target 92 Immuno-Oncology panel to characterize inflammatory protein dynamics associated with cytokine release syndrome (CRS). In the associated study, these proteomic data were integrated with wearable physiologic monitoring to evaluate early CRS detection and to identify candidate biomarkers, including IFN-γ.

Project description:Many cancer patients don’t benefit from currently-approved immune checkpoint inhibitors (ICI), suggesting that additional immunomodulation of the immunosuppressive tumour microenvironment (TME) is required. MTL-CEBPA specifically upregulates expression of master myeloid transcription factor, CEBPA, relieving myeloid-driven immunosuppression. Here, we report the safety, tolerability, pharmacokinetics, and efficacy of MTL-CEBPA in combination with pembrolizumab in patients with advanced solid tumours that typically show ICI resistance. Multimodal exploratory analyses of paired patient biopsies demonstrate biological changes associated with combination treatment of MTL-CEBPA and pembrolizumab, including increased infiltration of T cell and antigen-presenting cells supporting conversion from an immune desert towards a more immune-inflamed TME. Patients with disease stabilisation demonstrate reductions in immunosuppressive myeloid cells post-treatment. Collectively, these data support a role for MTL-CEBPA in reducing immunosuppression in the TME. This study was registered at ClinicalTrials.gov (NCT04105335). This manuscript also reports proteomic data from patients treated with MTL-CEBPA in combination with sorafenib from clinical trial, OUTREACH, accessible at ClinicalTrials.gov, number NCT02716012 and reported. Stored plasma from patients from this clinical trial were analysed using OLINK as described below.

Project description:The FIDO (Febrile Infant Diagnostic assessment and Outcome) study aimed to look at different diagnostic approaches to identify infants with a fever at low and high-risk of serious infection. FIDO was a multicentre prospective observational cohort study that enrolled infants ≤90 days of age presenting with fever to 35 Paediatric Emergency Research in the UK and Ireland (PERUKI) sites. The FIDO study was prospectively registered at ClinicalTrials.gov (NCT05259683; 28 February 2022). Where possible, residual plasma volume was collected and stored from infants for proteomic and biomarker analysis.

Plasma Proteome adaptations in Feminising Gender Affirming Hormone therapy and Pregnancy

Project description:The dataset consists of longitudinal plasma proteomic data was generated using the Olink proteomics platform in plasma from transgender individuals receiving feminizing gender-affirming hormone therapy (GAHT). There were two treatment arms in the clinical trial that received different anti-androgens to inhibit testosterone. This included a cyproterone acetate ("CPA") group (n=20) and a spironolactone ("SPIRO") group (n=20). Proteomics was measured in plasma at baseline and 6 months following feminizing GAHT. Additionally data in a cross-sectional pregnancy cohort at first and third trimester, as well as a cis female and cis male control were generated.

Proteomic Profiling of MIS-C Patients Reveals Heterogeneity R 1 elating to Interferon 2 Gamma Dysregulation and Vascular Endothelial Dysfunction

Project description:The pathologic mechanisms causing Multisystem 38 Inflammatory Syndrome in Children (MIS-C) are not yet known; we sought to use high throughput proteomics to uncover signatures of dysregulation suggestive of pathophysiology fo this condition. MIS-C is a major complication of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) pandemic in pediatric patients. Weeks after an often mild or asymptomatic initial infection with SARS-CoV-2 children may present with a severe shock-like picture and marked inflammation. Children with MIS-C present with varying degrees of cardiovascular and hyperinflammatory symptoms. We performed a comprehensive analysis of more than 1400 proteins in the plasma proteome of children with SARS-CoV-2 and MIS-C.

Evaluation of Olink Reveal Proximity Extension Assay for High-Throughput Proteomics: A Case Study Using NIST SRM 1950 and Spike-In Protein Standards

Project description:Plasma proteomics has regained attention in recent years through advancements in mass spectrometry instrumentation and sample preparation, as well as new high-throughput affinity-based technologies. Here, we evaluate the analytical performance of the new Olink Reveal platform, a proximity extension assay based technology quantifying 1,034 proteins across biological pathways. Using spiked-in recombinant Interleukin-10 (IL-10) and vascular endothelial growth factor D (VEGF-D) in the NIST SRM 1950 plasma standard, we assessed the linearity, sensitivity, precision and accuracy of the Olink assay. The results demonstrated strong linear relationships (R² 0.922–0.953) for both IL-10 and VEGF-D across spiked-in concentrations, confirming the robust technical performance of Olink Reveal and underscoring its suitability for relative quantitation in large-scale studies. The resulting data contains no sensitive or personally identifiable information, and is available in public repositories, and therefore suitable for use in benchmarking and software development.
