Project description:Canine oral melanoma is a highly aggressive tumor with limited therapeutic options and a poor prognosis. To elucidate the molecular mechanisms underlying its progression and response to radiation therapy (RT), we performed a longitudinal multi-omics analysis incorporating whole-genome sequencing (WGS) and RNA sequencing (RNA-seq) on tumor and blood samples from a single canine case. WGS was performed on tumor and blood collected before RT (Pre-RT) and on blood collected immediately after the final radiation fraction (Post-RT 0m). RNA-seq was performed on tumor and blood collected before RT (Pre-RT) and immediately after the final radiation fraction (Post-RT 0m), and on tumor collected 4 months after RT (Post-RT 4m).

Role of HSP70 in response to (thermo)radiotherapy: analysis of gene expression in canine osteosarcoma cells by RNA-seq

Project description:The aim of the study was to investigate which genes are up- and down-regulated in response to hyperthermia and combined thermoradiotherapy in HSP70 proficient and deficient canine osteosarcoma cell line. Canine osteosarcoma cell line Abrams was transfected with negative control siRNA or siRNA targeting HSP70. Cells were treated with hyperthermia (42C, 1 hour, HT), radiotherapy (6Gy, RT) and thermoradiotherapy (HTRT). RNA was extracted 24 hours after treatment and used for RNA sequencing. Three independent experiments were performed, each with negative control siRNA (Neg) and HSP70 knockdown (HSP70kd) cells. Cells were treated with radiotherapy (RT), hyperthermia (HT), thermoradiotherapy (HTRT) or untreated (ctrl). One experiment consists of 8 samples (ctrl, RT, HT, HTRT in neg and HSP70kd cells), total 24 samples were sequenced.

Project description:We have employed single cell RNA sequencing using the 10x Genomics platform to study the molecular mechanisms underlying resistance to treatment. In clinically relevant mouse models of G3 medulloblastoma, CT-guided fractionated radiotherapy was given to induce the clonal selection of radioresistant subpopulations. Comparison of recurrent and treatment naïve tumors revealed a prognostic gene expression signature reflective of radio-resistance.
