Exosome miRNA expression in umbilical cord blood of high-parity piglets reveals the reproductive potential of sows

Project description:The objective of modern pig breeding is to exhaust the genetic potential in reproduction performance of sows regarding to litter size and litter weight of piglets. During gestation period, umbilical cord contributes to placenta-fetal communication and plays an indispensable role in the intrauterine embryonic development. In this study, we attempted to analyze the molecular mechanism of reproductive declined in high-parity sows from the perspective of umbilical cord blood. Firstly, we analyzed the reproductive character data of sows, and then the histological analysis of umbilical cord phenotype was performed. Next, we evaluated the effect of umbilical cord blood exosomes (UCB-EXO) on angiogenesis. Moreover, the expression characteristics of miRNA in UCB-EXO of high-parity sows with poor reproductive performance (OS) and multiparous sows with excellent reproductive performance (MS) were analyzed. Results showed that the reproductive performance performed best at 3rd-7th and gradually decreased after 8th parities. Angiogenesis was repressed in OS piglets. Moreover, the Exo-MS exhibited pro-angiogenesis properties, with those of Exo-OS were diminished. With the increase of parities, the angiogenesis and immune function of sows decreased significantly, greatly limited the reproductive potential of sows. The data demonstrated that miRNAs of UCB-EXO played a central role in intrauterine development and suggested a novel possible explanation for reproductive potential, provides reference for increasing female reproductive efficiency.

Multi-omics analysis reveals maternal gut microbiota-derived short-chain fatty acids and progesterone are associated with offspring birth weight in sows

Project description:Piglet birth weight is a key determinant of preweaning survival and subsequent growth performance, yet the role of maternal gut microbiota in relation to offspring birth weight in sows remains incompletely characterized. This study aimed to investigate the association between maternal gut microbiota and offspring birth weight in sows. Fecal samples were collected from 260 Landrace × Yorkshire (LY) sows at gestation day 100, and sows were categorized into high birth weight (HBW; 1.41 ± 0.02 kg, 16.25 ± 0.25 piglets/litter, n = 59) and low birth weight (LBW; 1.07 ± 0.02 kg, 12.19 ± 0.22 piglets/litter, n = 52) groups based on the average birth weight of live-born piglets and live litter size. Fecal untargeted metabolomics was performed between groups.

Fermented sunflower meal in diets for Atlantic salmon

Project description:The increasing aquaculture production volumes have caused an escalating demand for alternative protein feed ingredients. Agro-industrial by-products such as sunflower meal are relatively abundant and cheap, but the inclusion levels are limited due to the presence of antinutritional factors and fiber. Solid state fermentation is a processing method with the aim of reducing the content of fiber and antinutritional factors and improving nutritional value in plant protein raw materials. In this study, Atlantic salmon (Salmo salar) at two commercial-like fish farms were fed diets containing 5% non-fermented sunflower meal (as a control diet) and two experimental diets with 5% or 10% fermented sunflower meal. The field trial lasted for eight and 11 months in a coastal and fjord location, respectively with the aim of comparing the effect of fermented diets on growth performance, gut microbiota, distal intestine histology and gene expression of Atlantic salmon under different environmental conditions. The findings revealed that diets with fermented sunflower meal sustained growth performance, improved intestinal health by reducing the prevalence of prominent inflammation and ectopic goblet cells and promoted gut lactic acid bacteria Lactiplantibacillus and Lactobaccilaceae after long-term feeding. Our results suggest that fermented sunflower meal is suitable as a protein source for Atlantic salmon when included at a level of up to 10% in the diet.

While both high fiber diet and N-Carbamylglutamate (NCG) have been individually demonstrated to enhance reproductive efficiency in sows, their potential synergistic effects and the involvement of uterine microbial mechanisms remain largely unexplored. In this study, we employed a multi-omics approach to investigate the synergistic impact of a high-fiber diet combined with 0.05% NCG on reproductive performance in Landrace × Yorkshire sows. The study revealed that high fiber with NCG (H+N) significantly increased FSH activity and T-AOC content in serum. Metagenomic analysis indicated an increased abundance of Clostridium disporicumin the uterine microbiota, which was positively correlated with elevated levels of the metabolite hydroxylysine. Subsequent transcriptomic profiling revealed activation of the PI3K-Akt signaling pathway, which was associated with increased FSH activity and T-AOC content. Overall, these findings demonstrated that the supplementation of a high-fiber diet with NCG enhances sow reproductive efficiency by modulating the uterine microbiota (specifically Clostridium disporicum), increasing hydroxylysine production, and activating the PI3K-Akt signaling pathway, thereby improving hormonal activity and antioxidant capacity.

Dietary High Fiber and N-Carbamylglutamate Enhance Sow Reproductive Performance via Modulating Lactobacilli, Lipid Metabolites, and the PI3K-Akt Signaling Pathway

Project description:Background Both high fiber diet and N-Carbamylglutamate (NCG) individually contributes to improve reproductive efficiency in sow. However, the potential synergistic effects and the underlying mechanisms are not well-understood. Here, Landrace x Yorkshire sows were selected to investigate the synergistic effects of high fiber diet and 0.05% NCG on reproductive performance using metagenomic, metabolomic, and transcriptomic analysis.Results The study revealed that high fiber with NCG (H+N) significantly increased litter weight at weaning and the apparent nutrient digestibility. Metagenomic analysis revealed H+N increased the abundance of lactobacilli across multiple taxonomic levels (from order to species level, including Lactobacillus sp. 910589175), which was positively correlated with elevated levels of the lipid metabolites L-α-glycerylphosphorylcholine and taurocholic acid. Subsequent transcriptomic profiling indicated activation of the PI3K-Akt signaling pathway, which was associated with improved litter weight at weaning and the apparent nutrient digestibility.Conclusion The synergistic effect of high-fiber diet and NCG supplementation enhances reproductive efficiency in sows by modulating gut microbiota (specifically lactobacilli), increasing beneficial lipid metabolites, and activating the PI3K-Akt signaling pathway, thereby improving nutrient digestibility and litter weight.
