To date few DDK substrates other than the MCM helicase have been identified, and none are implicated in fork protection or restart. Here we searched for such factors, using SILAC and quantitative mass spectrometry to identify the DDK phosphoproteome during fork stalling.

DNA repair by homologous recombination is under stringent cell cycle control. This includes the last step of the reaction, disentanglement of DNA joint molecules (JMs). Previous work has established that JMs resolving nucleases are activated specifically at the onset of mitosis. In case of budding y...

Initiation of eukaryotic DNA replication requires temporal separation of helicase loading from helicase activation and replisome assembly. Using an in vitro assay for eukaryotic origin-dependent replication initiation, we investigated the control of these events. After helicase loading, we found t...

Limonoid DDK restores mitochondrial redox homeostasis to alleviate metabolic dysfunction-associated steatohepatitis via promoting SDHA deacetylation and SDH complex activity
Mitochondrial dysfunction and redox imbalance are key drivers of metabolic dysfunction-associated steatohepatitis (MASH), yet therapeutic strategies targeting mitochondrial respiratory integrity remain limited. We identified succinate dehydrogenase subunit A (SDHA) as a direct functional target of t...
