Identification of SLBP-Interacting Proteins

ABSTRACT: To identify proteins that interact with SLBP in lung cancer cells, we performed immunopurification followed by mass spectrometry (IP-MS) in H1975 cells stably expressing Myc-tagged SLBP. Cellular extracts were subjected to affinity purification using anti-Myc magnetic beads, and the eluted protein complexes were resolved by SDS-PAGE and silver staining. Specific protein bands were excised and analyzed by mass spectrometry, revealing potential SLBP-binding partners.

Identification of SOX2 associated protein complexes in mouse ES cells

Project description:To uncover novel components of the SOX2-associated protein complex, we established E14 mouse embryonic stem (ES) cells stably expressing Flag-HA-tagged SOX2. Whole-cell extracts were prepared and subjected to tandem affinity purification, sequentially using anti-Flag M2 and anti-HA agarose resins . The purified SOX2 complex was separated by SDS-PAGE and visualized by silver staining . Distinct protein bands specifically enriched in the SOX2 immunoprecipitate were excised and analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) .

Immunoreactive proteins in the oesophageal gland cells of Anisakis simplex sensu stricto detected by MALDI-TOF/TOF analysis

Project description:Two hundred ventricles were manually dissected from L3 stage larvae of Aniskakis simplex s.s. to allow direct protein analysis. Denaturing gel electrophoresis followed by monochromatic silver staining revealed the presence of differential (enriched) proteins when compared to total nematode extracts. Such comparison was performed by means of 1D and 2D electrophoresis. Pooled antisera from Anisakis spp allergic patients were used in western blots revealing the presence of 13 immunoreactive bands in the ventricular extracts in 1D, while 82 spots in 2D. The corresponding protein bands and spots were excised from the silver stained gel and protein assignation was made by MALDI-TOF/TOF. A total of eleven proteins (including isoforms) were unambiguosly identified. The majority of these proteins are known to be secreted by nematodes into the external environment, of which three are described as being major allergens in other organisms with different phylogenetic origin.
