We evaluated eleven EV isolation methods to define their performance charac-teristics and optimal applications. EVs were quantified by NanoFCM, profiled for tetraspanins (CD9, CD63, CD81) via MSD assays, and further characterized by LC-MS/MS proteomics.

We developed a column-based CD9 antibody-immobilized HPLC immunoaffinity (CD9-HPLC-IAC) technology for EV isolation from a microliter-scale of serum for downstream proteomic analysis. The CD9-HPLC-IAC method achieved EV isolation from 40 μL of serum in 30 min with a yield of 8.0×109 EVs. Proteomic a...

To compare the efficiency, reproducibility, and proteomic profiles of extracellular vesicles (EVs) isolated from equine serum and plasma using size-exclusion chromatography (SEC) and differential ultracentrifugation (UC)+SEC, and to assess how the starting material influences EV cargo. Blood was col...

Growing evidence supports the importance of extracellular vesicle (EV) as mediators of communication in pathological processes, including those underlying respiratory disease. However, establishing methods for isolating and characterizing EVs remains challenging, particularly for respiratory samp...

The biological activity of extracellular vesicles (EVs) is largely defined by their molecular cargo, yet the impact of isolation workflows on EV proteomes and function remains incompletely understood. Here, we compared four isolation strategies for EVs derived from malignant ascites and ES-2 ovarian...

Secretome containing extracellular vesicles (EV) seem to mediate the benefits of cell therapy for ischemic heart failure. Our project has the objective of comparing the secretome containing extracellular vesicles (EV) from cardiac progenitor cells (EV-CPC) vs the secretome containing EV from Fibrobl...
