In this study, we characterised the interactomes of PAXX and its paralogs, XLF and XRCC4, to show that these factors share the ability to interact with DNA polymerase lambda (Poll), stimulate its activityand are required for the recruitment of Poll to laser-induced DNA damage sites.

Transcriptome profiling of PAXX knockdown in HCT116 colorectal cancer cells
To investigate the transcriptional changes induced by PAXX depletion in colorectal cancer cells, bulk RNA sequencing was performed in HCT116 cells transfected with PAXX-targeting siRNA or negative-control siRNA. Three independent biological replicates were analyzed for each group.
